摘要
目的探讨正常人外周血中TCRVα24+NKT细胞与传统CD3+T细胞在含量和亚群上的关系。方法采用五色荧光染色和流式细胞术检测TCRVα24+NKT细胞与传统CD3+T细胞在30例正常人外周血中所占比例,两群细胞中各亚群(CD4亚群、CD8亚群、双阴性亚群及CD45RA初始细胞亚群)在各自细胞群体中所占比例,统计软件分析两群细胞间各指标的差异及相关性。结果 NKT细胞在正常人外周血中所占比例为0.35%(0.10%~1.50%),CD3+T细胞比例为58.4%(43.2%~72.5%);在Va24+NKT细胞中CD4亚群所占比例为69.1%(21.3%~90.9%)、CD8亚群为15.0%(4.5%~76.2%)、双阴性亚群为9.5%(0.7%~69.4%)、CD45RA初始细胞亚群为32.4%(6.8%~54.0%),在CD3+T细胞中CD4亚群所占比例为54.7%(27.0%~74.7%)、CD8亚群为33.7%(19.9%~48.2%)、双阴性亚群为10.2%(1.9%~24.4%)、CD45RA初始细胞亚群为39.8%(10.3%~63.0%)。两群细胞间除CD4亚群和CD45RA亚群具有显著相关性(r=0.435 5,P<0.05;r=0.829 5,P<0.01),其余各指标均无相关性。CD4+NKT细胞含量与总NKT细胞含量呈显著负相关,而CD8+NKT与总NKT细胞呈显著正相关(r=-0.680 2,P<0.01;r=0.401 7,P<0.05)。结论正常人外周血中TCRVα24+NKT细胞与传统T细胞间除在CD4亚群和CD45RA初始细胞亚群间有显著相关外,其余各亚群相关较少。
Objective To examine the correlation between TCRVα24^+NKT cells and conventional T cells in terms of their frequency and proportion of subsets in peripheral bloods of normal persons.Methods Five-color fluorescence staining and flow cytometry were used to detect the frequency of TCRVα24^+NKT cells and conventional CD3+T cells in the peripheral blood of normal persons(n=30),and the proportions of CD4^+,CD8^+,double negative and CD45RA+subset in NKT cells and conventional CD3+T cells were measured as well.The difference and correlation between NKT cells and conventional CD3^+T cells were analyzed by using the statistical software.Results The frequency of NKT cells was 0.35%(0.10%-1.50%)and that of conventional T cells was 58.4%(43.2%-72.5%).The proportions of CD4+,CD8+,double negative and CD45RA+subset in NKT cells were 69.1%(21.3%-90.9%),15.0%(4.5%-76.2%),9.5%(0.7%-69.4%)and 32.4%(6.8%-54.0%),respectively,and those of each subset in conventional CD3^+T cells were 54.7%(27.0%-74.7%),33.7%(19.9%-48.2%),10.2%(1.9%- 24.4%)and 39.8%(10.3%- 63.0%),respectively.No correlations were found between TCRVα24+NKT cells and conventional CD3^+T cells except CD4 and CD45RA subsets between the two cells(r=0.435 5,P0.05;r =0.829 5,P0.01).There was an inverse correlation between the frequency of total NKT cells and the percentage of CD4^+NKT cells,while a positive correlation was noted between the frequency of NKT cells and the percentage of CD8+NKT cells(r=-0.680 2,P〈0.01;r=0.401 7,P〈0.05).Conclusion There was little correlation between TCRVα24+NKT cells and conventional T cells in peripheral bloods of normal persons except CD4+and CD45RA+subsets.
