摘要 目的建立美洲大蠊药材中尿嘧啶与肌苷含量测定的方法。方法采用HPLC法对尿嘧啶与肌苷的含量同时进行测定,色谱柱选用waters sunfire TM C18柱(4.6×250 mm;5μm),以3%甲醇溶液(含0.07%醋酸)为流动相A,甲醇为流动相B,流速为0.6 ml·min^-1,检测波长为254 nm,柱温为25℃。结果尿嘧啶对照品在0.020 3-0.202 8μg范围内与吸光度呈良好线性关系(r=0.999 96),加样回收率为99.90%(RSD=2.61%,n=6);肌苷对照品在0.103 76^-1.037 6μg范围内与吸光度呈良好线性关系(r=0.99999),加样回收率为99.94%(RSD=2.25%,n=6)。结论该方法操作简单,重复性好,精密度较高,可作为美洲大蠊中尿嘧啶与肌苷的含量测定方法。 Objective To establish a method for content determination of uracil and inosine in Periplaneta americana. Methods Content determination of uracil and inosine in Periplaneta americana by HPLC,the waters sunfire TM C18(4.6×250 mm; 5μm)was selected to be the chromatography column, with 3% aqueous methanol(containing 0.07% acetic acid) as the mobile phase A, methanol as mobile phase B, the flow rate is 0.6 ml·min^-1, the detection wavelength was 254 nm, the column temperature is 25 °C. Results The uracil in the range of 0.020 3 -0.202 8 μg(r = 0.999 96)was with a good linear relationship, and the total polysaccharide was 99.90%(RSD = 2.61%, n = 6); The inosine in the range of 0.103 76 - 1.03 76 μg(r = 0.999 99)was with a good linear relationship, and the total polysaccharide was 99.94%(RSD = 2.25%, n = 6). Conclusion The method is simple and can be used for the determination of uracil and inosine in Periplaneta americana.
