摘要
目的探讨三氧化二砷(arsenic trioxide,As2O3)对人脐静脉内皮细胞增殖活力的影响,评价As2O3防治经皮冠状动脉成形术(percutaneous transluminal coronary angioplasty,PTCA)后再狭窄的作用。方法将人脐静脉内皮细胞株(ECV-304)分为对照组和0.001、0.01、0.1、1μmol/L As2O3实验组,4个实验组细胞分别加入含0.001、0.01、0.1、1μmol/L As2O3的DMEM培养液,对照组加入等量不含As2O3的DMEM培养液。培养24、48、72、96h后进行活细胞计数,绘制细胞生长曲线,采用MTT试验观察As2O3对内皮细胞增殖活力的影响。结果细胞生长曲线显示As2O31μmol/L组内皮细胞增殖受抑制,0.001、0.01、0.1μmol/L各组无明显影响;1μmol/L组24、48、72、96h时细胞吸光度值(0.589±0.029、0.723±0.027、0.870±0.026、1.021±0.034)均低于对照组(0.628±0.027、0.898±0.045、1.118±0.031、1.118±0.031),差异有统计学意义(P<0.05);1μmol/L组96h时吸光度值高于72、48、24h时,72时高于48、24h时,48h时高于24h时(P<0.05)。结论 1μmol/L As2O3对内皮细胞增殖有抑制作用。
Objective To investigate the influence of arsenic trioxide(As2O3)on proliferation of human unbilical vein endothelial cells and evaluate its value to inhibiting restenosis after percutaneous transluminal coronary angioplasty(PTCA).Methods Human umbilical vein endothelial cells(ECV-304)were divided into the following five groups,0.001,0.01,0.1and 1μmol/L As2O3 groups and control group.The four experimental groups were incubated with DMEM of 0.001,0.01,0.1and 1μmol/L As2O3 respectively,and control group was incubated with equal amount of DMEM without As2O3.Cell survival number was described by cell growth curve and cell viability was measured by MTT method when the cells in five groups were cultured to 24,48,72 and 96hours.Results The cell growth curve showed that proliferation of endothelial cells was obviously inhibited in 1μmol/L As2O3 group,showing no significant differences among 0.001,0.01 and 0.1μmol/L As2O3 groups.The optical density values in 24,48,72 and 96hours were lower in1μmol/L As2O3group(0.589±0.029,0.723±0.027,0.870±0.026,1.021±0.034)than those in control group(0.628±0.027,0.898±0.045,1.118±0.031,1.118±0.031)(P0.05).In 1μmol/L As2O3 group,the optical density value was the highest in 96 hours,followed by that in 48 and 24hours(P0.05).Conclusion As2O3 of 1μmol/L inhibits the proliferation of endothelial cells.
