摘要
目的比较3种中性粒细胞碱性磷酸酶(NAP)染色方法的阳性率和阳性指数,选择一种染色效果好、技术范围宽的方法用于临床。方法根据NAP活性水平分为低、中、高组,选择61例高阳性组、100例正常阳性组、51例低阳性组;3组患者分别采用钙-钴法、未取代萘酚法、取代萘酚法3种方法染色,记录阳性率和阳性指数并作统计学分析。结果在各组中取代萘酚法阳性率和阳性指数最高,3种方法的阳性率和阳性指数比较有明显差异,P<0.05;两两比较3种方法阳性率和阳性指数,除低阳性组差异无统计学意义外(P>0.05),高阳性组和正常阳性组差异均有统计学意义(P<0.05)。结论取代萘酚法中性粒细胞碱性磷酸酶染色阳性率和阳性指数均优于其他2种方法,能够充分体现NAP的活性,在低阳性组也表达了低活性的NAP,减少了假阴性和假阳性,应首选在实验室推广应用。
Objective To compare three kinds tff neutrophil alkaline phosphatase (NAP) staining positive rate method and posi- tive index, select a method of good dyeing and wide range of techniques for the clinical departments. Methods According to the level of NAP activity, to group the low, mid, high, three kinds of cases, among of which 61 cases as high positive group, 100 cases as normal positive group, 51 cases as low positive group;The three groups were dyed respectively by calcium cobalt method, unsubstituted naphthol and substituted naphthol method, recording the positive rate and positive index and statistically analyze. Results The positive rate and the positive index of substituted naphthol method and positive index were the highest. The positive rate and positive index of three methods has significant differences, P 〈 0.05 ; comparing of the positive rate and positive index of every two methods between the three methods, there was no significant difference except the low positive group (P 〉 0.05). The high positive rate group and the nomal positive rate group both have statistical significance (P 〈 0.05). Con- clusion The positive rate and positive index of substituted naphthol neutrophil alkaline phosphatase staining method were better than the other two methods, can fully reflect the activity of NAP, in the low positive group also expressed the low activity of NAP, can reduce the false negative and false positive, and should be the first choice in the laboratory application.
