经腹腔镜子宫动脉闭塞后局部缺氧对子宫肌瘤细胞钙离子浓度变化的影响及其分子机制研究 被引量:4

摘要 目的:对比研究子宫肌瘤栓塞术后局部缺氧对子宫肌瘤细胞与周围平滑肌细胞钙离子浓度变化的差异及对细胞凋亡的调控机制。方法:比较20例子宫肌瘤患者在子宫动脉栓塞(UAOL)治疗前子宫肌瘤细胞及其周围平滑肌细胞IP3R1和RYR1的表达水平,并检测有/无钙通道阻断剂(肝素或/和钌红)下缺氧条件(1%O2)处理6小时后,两种细胞内Ca2+浓度,细胞色素C(Cytc表达水平及细胞凋亡情况。结果:患者子宫肌瘤细胞IP3R1和RYR1 mRNA和蛋白水平显著高于正常子宫肌层组织。在低氧条件下子宫肌瘤细胞内Ca2+浓度明显高于子宫平滑肌细胞[Ca2+:(82.69±16.92)nmol/L vs.(46.14±9.11)nmol/L,P<0.05];细胞色素C在两种细胞中增加量相同,钙离子阻断剂均可逆转上述子宫肌瘤细胞内Ca2+浓度,细胞色素C水平和细胞凋亡。结论:由IP3R1/RYR1介导的细胞内钙释放可诱导子宫纤维细胞在低氧条件下细胞凋亡,表明UAOL造成的缺氧环境可通过影响细胞内钙离子浓度造成子宫肌瘤细胞凋亡。
