乳腺癌中Sox17、β-连环蛋白和细胞周期蛋白D1的表达及其临床意义 被引量:4

Expression and clinical Implications of Sox17, β-catenin and Cyclin D1 in breast cancer

摘要 目的 探讨Sox17蛋白、β-连环蛋白(β-catenin)和细胞周期蛋白D1(Cyclin D1)在浸润性乳腺癌与良性乳腺增生组织中的表达及其临床意义.方法 应用免疫组织化学技术分别检测92例浸润性乳腺癌与60例良性乳腺增生组织中Sox17、β-catenin和Cyclin D1蛋白的表达,分析三者表达间的相关性及其与乳腺癌临床病理参数之间的关系.结果 乳腺癌组织中Sox17、β-catenin及Cyclin D1的阳性表达率分别为38.0% (35/92)、70.7% (65/92)、68.5% (63/92),与良性乳腺增生组织中的75.0% (45/60)、35.0% (21/60)和26.7% (16/60)间差异均有统计学意义(P<0.01);且Sox17的阳性表达与β-catenin和Cyclin D1的异常表达呈负相关(r=-0.675,P<0.01和r=-0.673,P<0.01),β-catenin与Cyclin D1间的异常表达呈正相关(r=0.693,P<0.01);Sox17表达阴性与肿瘤分期、淋巴结转移及组织学分级显著相关(P<0.01);β-catenin和Cyclin D1的异常表达均与肿瘤分期、淋巴结转移、雌激素受体(ER)及孕激素受体(PR)阳性显著相关(P<0.05).结论 Sox17在乳腺癌中表达降低与β-catenin和Cyclin D1表达增加显著相关,并且三者的表达与某些乳腺癌重要临床病理参数显著相关,提示三者在乳腺癌发生、发展过程中发挥重要作用. Objective This study aimed to investigate the expression and clinical significance of Sox17,β-catenin and Cyclin D1 in invasive breast cancer and benign breast disease.Methods Using immunohistochemical streptavidin avidin-peroxidase (SP) method,the expression of Sox17,β-catenin and Cyclin D1 was detected in 92 invasive breast cancer and 60 benign breast disease tissues.The correlations between Sox17,β-catenin and Cyclin D1 expression and corresponding clinicopathologic features were analyzed.Results The positive expression rates of Sox17,Cyclin D1 and β-catenin was 38.0% (35/92),70.7% (65/92) and 68.5% (63/92) in breast cancer tissues,which was significant different from 75.0% (45/60),35.0% (21/60) and 26.7% (16/60) in benign breast tissues respectively (P 〈 0.01).Correlation analysis showed that Sox17 protein expression was negatively correlated with the abnormal expression of β-catenin and Cyclin D1 (r =-0.675,P 〈 0.01 and r =-0.673,P 〈 0.01),while the abnormal expression of β-catenin protein was positively correlated with the expression of Cyclin D1 (r =0.693,P 〈 0.01).In clinically,negative expression of Sox17 protein was significantly correlated with tumor stage,lymph node metastasis and higher tumor grade (P 〈 0.01);the abnormal expression of β-catenin and Cyclin D1 were significantly correlated with tumor stage,lymph node metastasis,ER and PR receptors positive (P 〈 0.05).Conclusion The negative expression of Sox17 and the abnormal expression of β-catenin and Cyclin D1 are closely related in breast cancer,and they may be linked to various known clinicopathological features in breast cancer,suggesting that they may play an important role in the development and progression of breast cancer.
