基质金属蛋白-9及组织蛋白酶S在人股动脉粥样硬化斑块中的表达 被引量:4

Expression of matrix metalloproteinase-9 and cathepsin S in atherosclerotic plaques of human femoral arteries

摘要 目的 探讨基质金属蛋白酶-9(MMP-9)及组织蛋白酶S(CatS)在动脉粥样硬化斑块中的表达.方法 收集人股动脉的粥样硬化斑块组织45例,正常脾动脉组织24例,分为稳定斑块组(20例)、不稳定斑块组(25例)以及对照组(24例),采用免疫组织化学及反转录-聚合酶链反应(RT-PCR)检测各组标本中MMP-9和CatS及其mRNA的表达,以及两者间的关系.结果 MMP-9、CatS蛋白在平滑肌细胞、泡沫细胞、单核-巨噬细胞内有较多的表达,两者及其mRNA在各组中的表达差异有统计学意义(FMMP-9=168.448,P<0.01;FMMP-9 mRNA=221.300,P<0.01;FCats =273.246,P<0.01;FCats mRNA=51.681,P< 0.01).MMP-9、CatS蛋白及其mRNA在不稳定组(MMP-9蛋白:2.170±0.048,MMP-9 mRNA:1.588±0.085,CatS蛋白:2.231 ±0.050,CatS mRNA:1.411±1.104,P<0.05)、稳定组(MMP-9蛋白:2.080±0.051,MMP-9 mRNA:1.346±0.071,CatS蛋白:2.097±0.045,CatS mRNA:1.280±0.058,P<0.05)与对照组(MMP-9蛋白:1.959±0.011,MMP-9 mRNA:1.157 ±0.057,CatS蛋白:1.969 ±0.013,CatS mRNA:1.167±0.080,P<0.05)中的表达依次降低,两者及其mRNA在稳定组、不稳定组中的表达均呈正相关(r稳定蛋白=0.690,P<0.05;r稳定mRNA=0.739,P<0.01;r不稳定蛋白=0.940,P<0.01;r不稳定mRNA=0.820,P<0.01).结论 MMP-9、CatS蛋白的表达上调可能与动脉粥样硬化的发生及斑块的不稳定性有关. 0bjective To explore the expression of matrix metalloproteinase-9 (MMP-9) and cathepsin S (CatS) in antherosclerotic plaques.Methods Forty-five cases of human femoral arterial atherosclerotic plaques and 24 cases of nomal human splenic arterial samples (control group) were collected.The atherosclerotic plaques were divided into stable group (n =20) and unstable group (n =25).The expression levels of MMP-9 and CatS were detected by immunohistochemical staining and reverse transcriptase-polymerase chain reaction (RT-PCR).The correlations between MMP-9 and CatS were analyzed in stable and unstable groups.Results MMP-9 and CatS proteins were largely expressed in smooth muscle cells,foam cells and monocyte-macrophages.There was significant difference in the expression of MMP-9 and CatS protein and mRNA among groups (FMMP-9 =168.448,P〈 0.01;FMMP-9mRNA =221.300,P 〈0.01;FCatS =273.246,P 〈0.01;FCatS mRNA =51.681;P 〈0.01).The expression of MMP-9 and CatS protein and mRNA was reduced in unstable group (MMP-9 protein:2.170 ± 0.048,MMP-9mRNA:1.588 ±0.085,CatS protein:2.231 ±0.050,CatS mRNA:1.411 ± 1.104,P 〈0.05),followed by stable group (MMP-9 protein:2.080 ± 0.051,MMP-9 mRNA:1.346 ± 0.071,CatS protein:2.097 ±0.045,CatS mRNA:1.280 ±0.058,P 〈0.05),and control group (MMP-9 protein:1.959 ±0.011,MMP-9 mRNA:1.157 ±0.057,CatS protein:1.969 ±0.013,CatS mRNA:1.167 ±0.080,P 〈 0.05).There was a positive correlation between the expression of MMP-9 and CatS protein and mRNA in stable group and unstable group (rstable protein =0.690,P 〈 0.05;rstable mRNA =0.739,P 〈 0.01;runstableprotein=0.940,P〈0.01;rustablemRNA =0.820,P 〈0.01).Conclusion The enhanced expression of MMP-9 and CatS protein was possible associated with the formation of atherosclerosis and the instability of plaques.
