HPLC法测定儿茶配方颗粒中儿茶素和表儿茶素的含量

Content Determination of Catechin and Epicatechin in Catechu Dispensing Granules by HPLC

目的：建立同时测定儿茶配方颗粒中儿茶素和表儿茶素含量的方法。方法：采用高效液相色谱法。色谱柱为Shim-pack CLC-ODS-C18,流动相为0.04 mol/L枸橼酸溶液-N,N-二甲基甲酰胺-四氢呋喃（45∶8∶2,V/V/V）,流速为1.0 ml/min,柱温为35℃,检测波长为280 nm。结果：儿茶素和表儿茶素的进样量分别在0.301 6-1.508 0、0.202 0-1.010 0μg范围内与各自峰面积呈良好的线性关系（r=0.999 9、0.999 8）;精密度、稳定性、重复性试验的RSD均〈2%;平均加样回收率分别为96.60%（RSD=1.46%,n=9）、96.36%（RSD=1.30%,n=9）。结论：该方法简便、准确、重复性好,可用于测定儿茶配方颗粒中儿茶素和表儿茶素的含量。

OBJECTIVE：To establish a method for content determination Catechu dispensing granules.METHODS：HPLC was conducted with the column of Shim-pack CLC-ODS-C18.The mobile phase was 0.04 mol/L citric acid-N,N-two methyl formamide-tetrahydrofuran（45∶8∶2,V/V/V）,flow rate was 1.0 ml/min,column temperature was 35 ℃ and detecting wavelength was280 nm.RESULTS：There was a good linear relationship between the volume of catechin and peak area in the range of 0.301 6-1.508 0 μg（r=0.999 8）and epicatechin in the range of 0.202 0-1.010 0 μg（r=0.999 8）.The RSDs of precision,stability,repeatability test were less than 2%;the average recovery was 96.60%（RSD=1.46%,n=9）,96.36%（RSD=1.30%,n=9）.CONCLUSIONS：The method is simple,accurate and repeatable,and can be used for the content determination of catechin and epicatechin in Catechu dispensing granules.
