非那西丁和对乙酰氨基酚浓度评估肝脏储备功能

摘要 目的对非那西丁和对乙酰氨基酚浓度分析进行方法学研究,并进行应用性验证,以评估肝脏储备功能。方法建立高效液相色谱法(HPLC)测定非那西丁与对乙酰氨基酚浓度,进行标准曲线建立、精密度试验及方法回收率试验,并用健康人血浆进行应用性验证。结果非那西丁与对乙酰氨基酚在浓度为0.8-40.0μg/mL的范围内均呈现良好的线性关系。在5、15、25μg/mL样品浓度下,非那西丁测定的日内精密度分别为0.84%、1.09%和0.53%,日间精密度分别为1.51%、1.10%和1.38%;对乙酰氨基酚测定的日内精密度分别为0.76%、0.89%和0.67%,日间精密度分别为1.02%、1.46%和1.21%,在浓度为1-20μg/mL浓度范围内,利用所建立的分光光度技术测定,相对标准差(RSD)小于10%,当浓度超过5μg/mL,RSD〈5%。结论 HPLC简便快捷、灵敏度和准确度高,为评估肝脏储备功能提供了一种快捷、准确的方法。 Objective To perform the methodological study on detecting the levels of phenacetin and paraceta‐mol and to conduct the application verification for assessing the liver reserve function .Methods The high perform‐ance liquid chromatography (HPLC) method for detecting the levels of phenacetin and paracetamol was established . The standard curve was established .The precision and recovery tests were performed and the healthy plasma was a‐dopted for conducting the application verification .Results Both phenacetin and paracetamol showed a good linear re‐lationship in the concentration range of 0 .8 -40 μg/mL .The within‐day precisions of phenacetin under 5 ,15 ,25μg/mL samples were 0 .84% ,1 .09% and 0 .53% respectively ,which of paracetamol were 0 .76% ,0 .89% and 0 .67% ,respectively .The inter‐day precisions of phenacetin under 5 ,15 ,25 μg/mL samples were1 .51% ,1 .10% and 1 .38% ,which of paracetamol were 1 .02% ,1 .46% and 1 .21% ,respectively .RSD based on established spectrophoto‐metric techniques is less than 10% in the range of 1-20μg/mL Further ,It is less than 5% w hen the concentration is more than 5μg/mL .Conclusion The HPLC method is simple ,fast ,highly sensitive and highly accurate .It provides a fast and accurate method for assessing the liver reserve function .
