姜黄素抑制NF-κB信号对人肝癌HepG_2细胞株增殖的影响 被引量:10

摘要 目的探讨姜黄素对Hep G2细胞株增殖的影响及其机制。方法不同浓度的姜黄素(0、1.0、3.0、5.0、10.0、20.0、50.0、100.0μmol·L-1)处理Hep G2细胞株不同时间后,MTT法检测肝癌细胞株增殖情况;RT-PCR检测NF-κBp65和h TERT m RNA表达;Western blot检测h TERT蛋白表达,细胞免疫荧光法检测Hep G2肝癌细胞株中NF-κB活性。结果姜黄素能明显抑制Hep G2细胞的增殖,且呈时间与浓度依赖性;姜黄素能显著抑制NF-κBp65 m RNA表达及活性,同时伴有h TERT m RNA及蛋白表达降低。结论姜黄素抑制肝癌细胞株增殖,可能通过抑制NF-κBp65调控h TERT表达,进而下调端粒酶活性有关。 Objective To determine the regulatory effect of curcumin on NF-κB and hTERT gene in HepG2 cells, and to explore the antiproliferation mechanism of curcumin. Methods The expression of hTERT gene and NF-κB gene was analyzed by RT-PCR. Western blot was used to determine the protein expression of hTERT. NF-κB activity was measured with immunofluorescent microscopy. Results Curcumin inhibited NF-κB and hTERT expression. Conclusion Curcumin inhibits NF-κB activation and expression of hTERT in HepG2 cells in vitro. NF-κB may modulate hTERT at transcription level in HepG2 cells, and then cut the telomerase activity.
