白癜风患者血清中细胞因子和病变组织切除修复交叉互补蛋白1及γ-H2AX蛋白表达研究 被引量:1

The expression changes of cytokines, excision repair cross complementing-1 and γ-H2AX protein in pathological changes of vitiligo patients

摘要 目的 探讨白癜风患者血清中干扰素(IFN-γ)-γ2、白细胞介素(IL)-4、转化生子因子α(TGF-α)、肿瘤坏死因子-β(TNF-β)表达水平和病变组织中切除修复交叉互补蛋白(ERCC1)、γ-H2AX蛋白表达变化与白癜风分期的关系.方法 酶联免疫吸附试验(ELISA)检测不同分期白癜风患者血清中IFN-γ、IL-4、TGF-α、TNF-β表达水平的变化,蛋白印记(Western blot)检测病变组织中ERCC1和γ-H2AX蛋白表达变化.结果 稳定期和进展期白癜风患者与对照组相比,患者血清中IFN-γ和TGF-β表达水平降低(F=37.48,F=42.41,P<0.05),IL-4和TNF-β表达水平显著增加(F=56.37,F=77.19,P<0.05).进展期白癜风患者与对照组相比,患者血清的IFN-γ/IL-4比值降低(F=46.27,P<0.05);稳定期和进展期白癜风患者与对照组相比,病变组织中修复交叉互补蛋白(ERCC1)蛋白水平降低(F =45.71,P<0.05),而γ-H2AX蛋白水平显著增高(F =37.96,P<0.05).结论 IFN-y、IL-4、TGF-α和TNF-β参与白癜风的发病过程,ERCC1和γ-H2AX蛋白与白癜风病理损伤密切相关. Objective To explore the relationship between the expression of interferon-γ(IFN-γ),interleukin (IL)-4,transforming growth factor-α (TGF-α) and tumor necrosis factor-β(TNF-β) in serum and excision repair cross complementing-1 (ERCC1) and γ-H2AX protein in pathological tissues of vitiligo patients.Methods Enzyme-linked immunosorbent assay (ELISA)was used to detect the expression of IFN-γ,IL-4,TGF-α and TNF-β in serum of vitiligo patients with different phases.Western blot was used to detect the ERCC1 and γ-H2AX protein in pathological tissues of vitiligo patients.Results Compared with control,the expression of IFN-γ and TGF-β was decreased(F =37.48,F =42.41,P 〈 0.05),while the expression of IL-4 and TNF-β was increased in serum of vitiligo patients in stable and progressive phase(F =56.37,F =77.19,P 〈 0.05).Compared with control,the ratio of IFN-γ/IL-4 decreased in vitiligo patients in progressive phase (F =46.27,P 〈 0.05).Compared with control,the expression of excision repair cross complementing 1 ERCC1 protein was significantly decreased(F =45.71,P 〈 0.05),while γ-H2AX protein was significantly increased in pathological tissues of vitiligo patients in stable and progressive phase (F =37.96,P 〈 0.05).Conclusion IFN-γ,IL-4,TGF-α and TNF-β might take part in the pathogenesis of vitiligo.ERCC1 and γ-H2AX protein were associated with the pathological damage of vitiligo.
