摘要
为了改变现有的田间检测周期长以及受气候条件影响的弊端,建立一种高效、准确的纯度检测方法,采用SSR分子标记技术对黄瓜新品种‘津优401’种子的纯度鉴定结果进行比较。结果表明:在备选的100对引物中有12对引物在亲本间出现多态性互补的条带,尤其引物N67和N87,由于它们的特异性强、条带清晰的特点以及与田间鉴定的高度吻合被选为鉴定‘津优401’黄瓜新品种种子纯度的标记引物,从而将以往该品种种子纯度鉴定的时间从2-3个月缩短到4-5 h。
This study is used SSR markers technology, to explore the method of purity identification in new varieties of cucumber 'Jinyou 401' seed, the main research goal is to establish an efficient, accurate purity identification method, to solve the shortcomings of the existing methods, such as, shorten the identification cycle and weaken the climate conditions effect. The results showed that the alternative 100 pairs of primers have 12 pairs of primers complementary to appear polymorphic bands between the parents, particularly the primer N67 and N87, because of their specificity, as well as the characteristics of clear bands and the anastomosis with field test, was chosen the labeled primers, to identify ' Jinyou 401', which will shorten the seed identification time from 2-3 month to 4-5 hours.
