摘要
为探索新材料,降低昆虫刺吸电位技术(Electrical Penetration Graph,EPG)的测试成本,本研究以镀银碳纤维为材料,以金丝为对照,在直流刺吸电位仪(DC-EPG)上测试Q型烟粉虱Bemisia tabaci的取食行为,并对波形进行分析比较。结果表明,利用镀银碳纤维记录的EPG波形总体上与金丝的非常类似;分析几类典型波形可以发现,与金丝相比,镀银碳纤维记录的pd波相对振幅显著下降,且pd-Ⅱ亚波的Ⅱ-1和Ⅱ-3部分的频率显著增大;E2波电压水平显著低于金丝记录的水平,而两者记录韧皮部参数的相对振幅和频率以及E1波的电压水平差异均不显著。说明镀银碳纤维在一定程度上能够改进EPG的记录结果,提高波形的精确性,可以在EPG实验中替代金丝,但在导电性方面尚有改进的空间。
In order to explore possible application of the cheaper,new material in Electrical Penetration Graph( EPG) experiments,in this study,waveforms of Q biotype Bemisia tabaci feeding behavior in direct-current electrical penetration graph( DC-EPG) were recorded and compared by parallel using of the silver-plated carbon fiber and the golden wire. The results showed that the EPG-recorded waveforms with silver-plated carbon fiber were almost similar to those with golden wire. By comparatively analyzing typical waveforms,it is found that the waveforms with silver-plated carbon fiber had lower relative amplitude than those with the golden wire. Ⅱ-1 and Ⅱ-3,sub-waveforms of waveform potential drops( pd),differently higher frequency was observed. The waveform E2 in recordings using silverplated carbon fiber had significantly lower voltage levels compared with those recordings with golden wire,but for waveform E1,there were no significant differences in relative amplitudes,frequencies and the voltage levels between both recordings. The results indicated that silver-plated carbon fiber could be used to replace gold wire in the EPG recordings to obtain more precise waveforms,but there is still some space for improvement of the electrical conductivity.
