咳喘停贴剂穴位贴敷治疗对慢性哮喘大鼠肺组织形态学的影响

Effects of Acupiont Sticking Therapy with Kechuanting Paste on Morphology of Lung in Chronic Asthma Rats

目的:观察咳喘停贴剂穴位贴敷治疗对慢性哮喘大鼠肺组织光镜及电镜下形态学的影响。方法:将40只wistar大鼠,随机分成正常组(A)、模型组(B)、假穴位贴敷组(C)、地塞米松组(D)、穴位贴敷组(E)。B、C、D、E组以卵蛋白(OVA)致敏和激发制作慢性哮喘模型。观察治疗后各组大鼠光镜下肺组织形态学改变及电镜下肺泡、肺间质超微结构的变化。结果:光镜下:A组肺泡及肺间质结构正常;B组与C组相似,肺泡内见有炎性细胞浸润,肺间隔增厚。D组及E组:与模型组相比,炎性细胞浸润呈不同程度的减少,肺间隔增生呈不同程度的减轻。电镜下:A组见到形态良好的肺泡Ⅱ型上皮细胞。B组及C组见肺泡Ⅱ型上皮细胞板层小体排空现象较多,细胞表面微绒毛明显减少,间质内见大量胶原纤维增生及浸润的淋巴细胞。D组及E组见Ⅱ型肺泡上皮细胞结构基本正常,肺间质内可见淋巴细胞及束状胶原纤维。结论:咳喘停贴剂可通过影响肺泡及肺间质的形态来影响其功能,从而达到治疗哮喘的目的。

Objective： To observe the effects of acupoint sticking therapy with Kechuanting paste on morphology of lung in chronic asthma rats with light microscopy and electron microscopy. Method： The 40 wistar rats were randomly divided into nor- mal group （A）, model group （B）, sham acupoint application group （C）, dexamethasone （D） and acupoint application group （E）. B, C, D, E groups all received ovaIbumin （OVA） to establish the model of chronic asthma. Morphological changes under light mi- croscopy and alveoli, interstitial lung ultra-structural changes under electron microscopy were observed after the treatment. Re- suits： Light microscope： the structure of alveolar and interstitial lung in A group was normal; the situation of group B and C is similar, there are inflammatory cells in alveolus, and pulmonary septal thickening. Group D and E： Compared with the model group, the number of inflammatory cells decreased differently, lung hyperplasia interval showed varying degrees of ease. Electron microscope： The type Ⅱ cells of alveolar in A group showed good shape. Emptying phenomenon of lamellar body in type II ep- ithelial cell increased in group B and C, micro-villi of cell surface decreased significantly, and a large number of collagen fibers and lymphocytes appeared. The structure of type Ⅱ alveolar （epithelial） ceils in group D and E was normal, and there were lymphocytes and bundles of collagen fibers. Conclusion： Kechuanting Paste can improve the lung function through affecting the morphology of the alveolar and interstitial lung, to cure asthma.
