响应面法优化野葛叶中叶绿素超声波提取工艺 被引量:10

摘要 为优化野葛叶叶绿素提取工艺,在超声波辅助的基础上,选取影响叶绿素提取效果的液固比、超声提取温度、超声提取时间、提取溶剂种类四个因素,在单因素实验的基础上,根据Box-behnken中心组合设计原理,以叶绿素得率为影响值,进行四因素三水平的响应面分析(RSA)。结果表明,超声波辅助提取野葛叶中叶绿素优化条件为:液固比106∶1(m L·g-1),超声提取温度54℃,超声提取时间31min,提取溶剂种类乙醇∶丙酮(1.9∶1),在此条件下,叶绿素得率可达0.691%。 In order to optimize the ultrasound extract of Pueraria Iobata leaves chlorophyll process conditions, four factors were selected which had effect on extraction rate of chlorophyll, that was ratio of liquid to material, ultrasonic extraction temperature, ultrasonic extraction time,extraction solvent species. In the basis of single factor experiments,these four factors were optimized using response surface analysis based on a four-variable,three- level Box-Behnken design.The results showed that the optimum preparation technology for ultrasound extract of Pueraria Iobata leaves chlorophyll were. ratio of liquid to material was 106:1 (mL·g^-1), ultrasonic extraction temperature was 54℃, ultrasonic extraction time was 31min, extraction solvent species ethanol: acetone (1.9:1 ). Under the optimal extraction condition,the chlorophyll productivity was 0.691%.
