摘要
目的明确沉默ABCG2后膀胱癌T24对X线的敏感性是否增强。方法常规培养T24,经4 Gy X线照射后,从存活能力、克隆形成、迁移和侵袭能力四个方面评价了沉默ABCG2后T24对X线敏感性的变化。结果经X线照射,沉默ABCG2后,T24的存活能力、克隆形成、迁移、侵袭能力显著减弱(P<0.05)。结论沉默ABCG2增强了膀胱癌T24对X线的敏感性,ABCG2的抑制剂可能成为膀胱癌的放疗增敏剂。
Objective To investigate the roles of ABCG2 in radiotherapy sensitivity of bladder transi- tional cell cancer cell line T24. Methods Three pairs of siRNA was designed to target 5'-UTR of ABCG2 mR- NA to decrease its expression through mRNA degradation. Western blot was used to test silence efficiency. MTT, plate colony forming experiment, transwell migration and invasion experiment were used to test survival, proliferation ,migration and invasion ability. X-ray was used to test radiotherapy sensitivity. Results Western blot assay suggested siRNA 3# has biggest silence efficiency. After silence of ABCG2 and X-ray treatment, the survival,proliferation, migration and invasion ability of T24 was decreased significen lence of ABCG2. Conclusion Silence of ABCG2 increased X,ray sensitivity of T24. aradiotherapy sensitivity enhancer. fly compared with no si- ABCG2 inhibitor maybe
