摘要
目的探讨细胞周期相关激酶(CCRK)在大肠癌组织中的表达及其与大肠癌临床病理特征之间的关系。方法选取50例经病理学确诊的原发性大肠癌患者的癌组织,以配对的癌旁组织作为对照,采用RT-PCR和Western blot检测CCRK mRNA和CCRK蛋白表达的差异性。结果 CCRK蛋白在大肠癌组织中的阳性率为60.00%(30/50),癌旁结肠组织阳性率为40.00%(20/50)差异有统计学意义(χ2=5.769,P<0.05);CCRK蛋白在大肠癌组织中的相对表达量为2.69±0.87,癌旁组织为1.00±0.28,差异有统计学意义(t=5.81,P<0.05)。CCRK mRNA在大肠癌组织中的阳性率为64.00%,癌旁组织为42.00%,差异有统计学意义(χ2=4.857,P<0.05);CCRK mRNA在大肠癌组织中的相对表达量为2.45±0.92,癌旁组织为1.00±0.21,差异有统计学意义(t=4.83,P<0.05)。PT3-4期大肠癌组织CCRK mRNA阳性率为85.70%(12/14),PT1-2期大肠癌组织阳性率55.56%(20/36),差异有统计学意义(χ2=3.979,P<0.05);PN1-2期大肠癌组织CCRK mRNA阳性率为87.50%(14/16),PN0期为52.94%(18/34),差异有统计学意义(χ2=5.640,P<0.05)。结论 CCRK的高表达与大肠癌发生、发展及其TNM分期有关,检测CCRK蛋白和CCRK mRNA表达可以作为大肠癌早期诊断和预后的一项重要参考指标。
Objective To examine the expression of a cell cycle-related kinase(CCRK)in colorectal cancer tissue and its relationship to the clinicopathological features of colorectal cancer. Methods Subjects were 50 patients who were confirmed by pathology to have primary colorectal cancer.Cancerous tissue from these patients was compared to matching tissue adjacent to the carcinoma.RT-PCR and Western blotting were used to examine differences in expression of CCRK mRNA and protein. Results Colorectal cancer tissue tested positive for expression of the CCRK protein at a rate of 60.00%(30/50)while paracancerous tissue tested positive for expression of the CCRK protein at a rate of(40.00%).The rate of protein expression in those tissues differed significantly(χ2= 5.769,P〈0.05).The relative level of CCRK protein expression in colorectal cancer tissue(2.69±0.87)was significantly higher than that in paracancerous tissue(1.00±0.28,t=5.81,P〈0.05).The rate at which colorectal cancer tissue tested positive for expression of CCRK mRNA(64%)was significantly higher than the rate at which paracancerous tissue tested positive for expression of CCRK mRNA(42%,χ2=4.857,P〈0.05).The relative level of CCRK mRNA expression in colorectal cancer tissue(2.45±0.92)was significantly higher than that in paracancerous tissue(1.00±0.21,t=4.83,P〈0.05).The rate at which PT3-4stage colorectal cancer tissue tested positive for expression of CCRK mRNA(85.7%)was significantly higher than the rate at which PT1-2stage colorectal cancer tissue tested positive for expression of CCRK mRNA(55.6%,χ2=3.979,P〈0.05).The rate at which PN1-2stage colorectal cancer tissue tested positive for CCRK mRNA expression(87.5%)was significantly higher than the rate at which PN0 stage colorectal cancer tissue tested positive for CCRK mRNA expression(52.9%,χ2=5.640,P〈0.05). Conclusion A high level of CCRK expression was associated with the occurrence of colorectal cancer and its TNM stage.Monitoring the expression of the CCRK protein and CCRK mRNA has the potential to be an important indicator for the early diagnosis of colorectal cancer and patient prognosis.
