P16基因与细胞衰老 被引量:1

摘要 细胞衰老,最初被描述为细胞老化的一种形式,是指细胞在信号转导作用下不可逆地脱离细胞周期并丧失增殖能力后进入的一种相对稳定的状态。细胞衰老的过程与多种刺激或应激原触发有关,包括癌基因的激活,端粒的损耗缩短,氧化应激,DNA 损伤,病毒感染,紫外线辐射等等[1,2]。与衰老相关的基因有很多种,但关键基因的作用不可或缺, P16基因既是一种抑癌基因,又被认为是细胞衰老的主导基因,对细胞衰老的启动及维持均具有重要的作用[3]。研究发现,在细胞衰老过程中,P16水平上调可达几十倍,甚至上百倍,P16基因在衰老细胞中处于高表达水平[4,5]。高表达的 P16与细胞周期依赖性激酶形成复合体,导致细胞周期阻滞于 G1期(DNA 合成前期),不能进入细胞周期循环而处于静息状态。P16基因是目前细胞衰老研究的热门靶点之一。现就 P16基因与细胞衰老的关系作如下综述。
