摘要 利用固定金属离子亲和色谱法即IMAC(Immobilized Metal-ion affinity chromatography)的原理,使用表面修饰Ni2+离子的金纳米粒子对牛血红蛋白的吸附和脱附行为进行了研究。粒子对牛血红蛋白的最大吸附量可达到5.56mg/mg。粒子可以将牛血红蛋白从牛血红蛋白和牛血清蛋白混合物中分离出来。通过紫外光谱及电泳测试进一步论证了前面的结论。这种方法简单、有效、方便,在牛血红蛋白的分离纯化方面有着很好的应用前景。 The adsorption behavior of bovine hemoglobin on Ni2+immobilized gold nanorparticles was investiga-ted,which refer to the method of Immobilized Metal-ion affinity chromatography ( IMAC) . The particles showed high absorption efficiency to bovine hemoglobin and the maximum extraction amounts reached 5. 56mg/mg. The particles were applied to separate bovine hemoglobin from the mixture of bovine hemoglobin and bovine serum albumin. These results were proved by UV-vis spectra and Protein Gel Electrophoresis. This method is a sim-ple,convenient and effective way,which has potential application in removing abundant protein in proteomic a-nalysis.
