rhBNP对慢性心力衰竭兔心功能的影响及机制

摘要 目的探讨重组人脑利钠肽(rh BNP)对慢性心力衰竭(HF)模型兔心功能的影响及可能的机制。方法以阿霉素诱导HF家兔模型(24只),随机分为对照组、模型组、治疗组。连续治疗4周后,测试左心功能,采用酶联免疫吸附法测定血清中AngⅡ的含量,应用RT-PCR检测心肌Ⅰ、Ⅲ型胶原的mRNA表达情况。结果治疗组左心室结构和收缩功能各项指标显著优于模型组,差异有统计学意义(P<0.05);模型组心肌Ⅰ、Ⅲ型胶原含量明显高于正常对照组,差异有统计学意义(P<0.05);与模型组比较,治疗组心肌Ⅰ、Ⅲ型胶原含量的表达下降,差异有统计学意义(P<0.05);治疗组血清中AngⅡ含量显著低于模型组,差异有统计学意义(P<0.05)。结论 rh BNP能抑制HF家兔心肌中Ⅰ、Ⅲ型胶原含量表达,一定程度上能拮抗血管紧张素Ⅱ(AngⅡ)的活性,改善心室重塑,缓解HF症状。 Objective To observe the effect and explore the possible mechanism of rhBNP on cardiac function of heart failure rabbit model. Methods Heart failure rabbit models ( n=24 ) were induced by adriamycin. 24 rabbits were randomly divided into control group, model group and treatment group. After 4 weeks treatment, the left heart function was tested,and the plasma angiotensinⅡcontent was determined by ELISA,the mRNA expression in collagenⅠand collagen Ⅲ was detected by RT-PCR. Results The indexes of left ventricular structure and systolic function in treatment group was significantly better than that of model group (P〈0. 05);The myocardial collagenⅠand collagenⅢ content of model group was obviously higher than that of control group (P〈0. 05);Compared with model group, the expression of collagen Ⅰ and collagen Ⅲ in treatment group decreased significantly ( P〈0. 05 ); The serum AngⅡ content in treatment group was significantly lower than that of model group ( P〈0. 05 ) . Conclusion rhBNP can inhibit the expression of collagenⅠand collagen Ⅲ in heart failure rabbits, antagonize the activity of angiotensin Ⅱ( Ang Ⅱ) in a certain extent,and improve the ventricle remodeling.
