依达拉奉对大鼠全脑缺血再灌注损伤的保护作用 被引量:2

The protection of edaravone on global cerebral ischemic-reperfusion injury in rats

摘要 目的探讨依达拉奉对大鼠全脑缺血再灌注损伤模型运动功能的影响,并进一步对脑组织含水量、丙二醛(malondialdehyde,MDA)、NO含量及超氧化物歧化酶(superoxide dismutase,SOD)活性进行测定,观察依达拉奉对全脑缺血再灌注损伤的保护作用。方法选择2014年8—11月清洁级健康雄性SD实验大鼠45只,随机分为假手术组(A组)、缺血再灌注组(B组)、依达拉奉组(C组);术前5 min静脉注射0.9%氯化钠溶液,1 mg/kg,全脑缺血再灌注后24 h对实验大鼠进行运动功能评估,测定各组大鼠脑组织含水量,并检测脑组织MDA、NO含量和SOD活性,计量资料以x±s表示,采用t检验,P<0.05为差异有统计学意义。结果 B组和C组大鼠平衡评分结果[(1.52±0.92)、(3.02±0.71)分]和引体试验评分结果[(1.07±0.83)、(2.26±0.77)分]与A组平衡及引体试验评分结果[(4.27±0.69)、(4.03±0.65)分]比较均下降,差异均有统计学意义(均P<0.05);B、C两组比较,C组平衡试验与引体试验评分升高,差异均有统计学意义(均P<0.05)。B组与C组两组实验大鼠脑组织含水量分别为(79.02±0.65)%、(77.85±0.59)%,较A组的(75.31±0.41)%明显升高,差异均有统计学意义(均P<0.05);与A组[(0.33±0.12)μmol/mg、(6.02±0.21)nmol/mg、(76.55±2.64)U/mg]比较,B组和C组NO、MDA含量[(0.96±0.39)、(0.63±0.06)μmol/mg、(7.75±0.49)、(6.97±0.40)nmol/mg]明显升高,SOD活性[(40.07±3.23)、(53.25±4.57)U/mg]显著降低,差异均有统计学意义(均P<0.05);C组较B组可显著降低全脑缺血再灌注损伤后MDA、NO含量,提高SOD活性,差异均有统计学意义(均P<0.05)。结论依达拉奉预处理对全脑缺血再灌注损伤所致运动功能障碍,有一定程度的改善;并可降低MDA及NO水平,而显著提高SOD活力,对大鼠全脑缺血再灌注损伤后的脑组织有一定的保护作用,能显著改善预后。 Objective To investigate the effect of edaravone on global cerebral ischemic-reperfusion injury by evaluating the motor function,detecting the brain water content, measuring malondialdehyde(MDA),NO content and superoxide dismutase(SOD) activity in brain tissue.Methods 45 male SD rats were randomly divided into sham operation group(group A),ischemic-reperfusion control group(group B) and edaravone group(group C) from August to Novemeber in 2014.1 mg/kg of 0.9% sodium chloride Solution was intravenous administration preoperative 5 rain.The motor activity tests were carried out 24 h after reperfusion.Then the rats were killed,the brain water content was detected and brain tissues were taken for determination of the concentration of MDA,NO and activity of SOD.Count data were processed by t-test,P〈0.05 was considered statistically significant.Results Compared with group A[(4.27 ± 0.69),(4.03 ±0.65)points],the motor activity were significantly descending in groups B[(1.52 ±0.92),(3.02 ± 0.71) points]and C [(1.07 ± 0.83),(2.26 ± 0.77)points](all P〈0.05).It can be found the motor activity scores of group C is higher than the control group(P〈0.05).Compared with group A[(75,31 ± 0.41)%],the water content of brain were significantly improved in groups B [(79.02± 0.65)%] and C [(77.85 ± 0.59)%](all P〈0.05).Compared with group A [(0.33 ± 0.12)μmol/mg, (6.02± 0.21)nmol/mg, (76.55 ± 2.64)U/mg],the contents of NO and MDA [(0.96 ±0.39), (0.63 ± 0.06)μmol/mg, (7.75± 0.49), (6.97 ± 0.40)nmol/mg] were significantly improved and activitives of SOD [(40.07± 3.23),(53.25 ± 4.57)U/mg] were decreased in groups B and C(all P〈0.05). It was found that contents of NO and MDA in group C than B were significantly decreased,SOD activities was significantly increased(all P〈0.05).Conchmion Edaravone can improve the motor function and the antioxidant capacity of brain tissue,which has a neuroprotective effect against global cerebral ischemia-reperfusion injury in rats.
