槐米及其不同成分对内皮细胞损伤修复作用比较研究

Protective Effects of Different Components in Sophorae flos on High Glucose-induced Injury in Human Umbilical Vein Endothelial Cell

目的:比较研究芦丁、槲皮素、槐米黄酮部位和槐米药材提取物对高糖诱导的内皮细胞损伤的修复作用。方法:采用含有33mmol·L-1葡萄糖和1%FBS的DMEM低糖培养基处理人脐静脉HUVEC内皮细胞,建立细胞损伤模型,采用NO试剂盒检测各样品对模型内皮细胞NO分泌量的影响;采用MTT法测定细胞活力。结果:芦丁、槲皮素、槐米黄酮部位和槐米药材提取物均可不同程度地升高模型HUVEC内皮细胞NO分泌量,不同程度地增强细胞活力。结论:芦丁和槲皮素可能为槐米修复内皮细胞损伤的活性成分。

Objective;To evaluate the protective effects of rutin, quercetin,Flavonoids compounds of Sophorae flos, extract of Sophorae flos on the injured human umbilical vein endothelial cell. Methods.. High glucose-induced injury in human umbilical vein endothelial cell, determine cell survival viability by MTT, determine the NO produced in cell supernatant by means of nitrate reduetasemethod. Results：A certain concentration of rutin, quercetin,Flavonoids compounds of Sophorae flos, extract of Sophorae flos can enhance the activity of cell under high glucose condition, inhibit the recession of NO. Conclusion： Rutin and quereetin could be the active ingredient of Sophorae flos.
