小花棘豆中毒对小鼠脑组织α-甘露糖苷酶表达的影响 被引量:3

摘要 探讨小花棘豆对小鼠脑组织α-甘露糖苷酶(AMA)表达的影响,进一步揭示小花棘豆的毒性作用机理。将40只小鼠随机分为4组,即对照组、试验Ⅰ组、试验Ⅱ组和试验Ⅲ组,3个试验组分别按照每千克体重1、5、10 g的剂量饲喂小花棘豆,攻毒后第63天采集小鼠脑组织,检测小鼠不同脑区AMA分布与表达的变化。结果显示,小花棘豆中毒可导致小鼠脑组织AMA mRNA表达水平的下降,但试验Ⅰ组与对照组差异不显著(P>0.05),试验Ⅱ组和试验Ⅲ组小鼠小脑、大脑和丘脑AMA mRNA表达量均显著低于对照组(P<0.05),但3个试验组海马和脑干AMA mRNA表达量与对照差异均不显著(P>0.05)。AMA在海马与脑干中表达极弱,在其他区域均有表达,表达量与神经细胞和颗粒细胞的数量正相关。结果表明,不同剂量小花棘豆可降低小鼠脑组织AMA mRNA表达量,小脑、大脑和丘脑是其主要作用的靶区。
