摘要
核因子E2相关因子2是重要的氧化还原调节剂。基础条件下,核因子E2相关因子2位于细胞浆,与抑制因子Keap1结合而降解;在应激因素作用下,核因子E2相关因子2激活并与Keap1解离,转位于细胞核内,与多种抗氧化应激基因和Ⅱ相解毒酶的抗氧化反应元件结合,促进血红素氧化酶1等抗氧化应激蛋白和多种解毒酶的表达增加,进而发挥抗氧化应激调节作用。研究显示,核因子E2相关因子2及其靶基因激活在保护内皮功能、血管生成、心肌再生和支架内再狭窄等方面发挥重要作用。现综述核因子E2相关因子2与心血管疾病相关研究的新进展。
Nuclear factor E2-related factor 2 ( Nrf2) is an important redox regulator.Under basal conditions, Nrf2 is localized in the cytoplasm where it binds to the Kelch like ECH associating protein (Keap1) and is degraded by the proteasome pathway.Once the cells are exposed to various oxidative or electrophilic inducers, Nrf2 is dissociated from Keap1 and translocates to the nucleus where, by binding to the anti-oxidant response element in the promoters of many anti-oxidant genes such as heme oxygenase-1, and it induces their expression.It has been reported that Nrf2 and its target proteins play critical roles in the protection of endothelial function, angiogenesis, regeneration of cardiac cells and in-stent restenosis and so on.In this paper, the progress in identifying the relationship between Nrf2 and cardiovascular disease are reviewed.
