摘要
以六株具有石油降解功能的细菌(编号为GW40、GW101、GW109、GW117、GW157、GW159)为材料,通过全细胞蛋白电泳聚类,16S r RNA基因系统发育分析和生理生化特性初步鉴定,鉴定结果为不动杆菌属(Pseudomonas sp.)。并研究了不同p H,温度和石油浓度对菌株降解石油能力的影响。SDS-PAGE图谱分析结果表明,在14.4—96.4KDa范围内,它们有92.3%以上相似性蛋白图谱,证明它们属于同一类群;16S r RNA基因系统发育分析表明六株菌与不动杆菌属的亲缘关系较近,因此把它们归到不动杆菌属。六菌株在p H 6.0—7.0,温度为30℃和石油浓度为2 g·L-1时,对石油的降解率最大。
Diversities of six petroleum-degrading strains GW40, GW101, GW109, GWll7, GW157 and GW159 were analyzed by SDS-PAGE of the whole cell, morphological and biochemical characters and 16S rRNA gene sequence phylogenetic analysis. The patterns of SDS-PAGE of the whole cell show that six strains have high similarity (more than 92.3%) in the range of 14.4-94 kDa. 16S rRNA gene sequence phylogenetic analysis and morphological and biochemical characters indicate six strains have higher similarity with species of the genus Acinetobacter and were allocated to the genus Acinetobacter. Six strains have higher degradation ratio at pH 6.0-7.0, a temperature of 30 ℃ and the petroleum concentra- tion of 2 g·L-1.
