HER-2蛋白表达和基因扩增状态在乳腺癌中的一致性研究 被引量:2

摘要 目的探讨乳腺癌HER-2蛋白表达和基因扩增状态的一致性关系。方法采用免疫组化法和荧光原位杂交法检测120例乳腺癌组织中HER-2蛋白表达和HER-2基因扩增情况,分析乳腺癌中2种监测方法的一致性及与临床病理参数的关系。结果乳腺癌组织中HER-2蛋白表达阳性率为70.83%,而基因扩增率为19.16%,在85例HER-2蛋白表达阳性组织中有21例HER-2基因扩增阳性,而在23例HER-2基因扩增阳性的乳腺癌组织中仅2例无HER-2蛋白的过表达,HER-2蛋白表达和基因扩增间具有显著相关性(P<0.01)。HER-2蛋白的表达和HER-2基因扩增均与乳腺癌病灶大小和组织学分级密切相关(P<0.05),而与患者年龄、肿瘤的临床分期、腋窝淋巴结转移及ER和PR的状态无明显相关性(P>0.05)。结论乳腺癌组织中HER-2蛋白表达和基因扩增与肿瘤分化程度密切相关,两者具有较好的一致性。 Objective To investigate the consistency about HER -2 protein expression and HER -2 gene amplification state in breast cancer tissue. Methods The expression of HER - 2 protein and HER - 2 gene were detected with immunohistochemical and fluorescence in situ respectively in 120 cases of breast cancer tissue. The consistency about them in breast cancer and its re- lationship with clinicopathological parametersits were analyzed. Results The HER -2 protein and gene amplification expres- sion positive rate in breast cancer tissue was 70.83% and 19.16%, respectively. 85 cases tissue being with HER -2 protein positive expression had 23 cases of HER - 2 gene amplification positive in breast cancer tissues. But 23 cases issue being with HER- 2 gene positive expression without HER - 2 protein expression in 2 cases only. It had significant correlation between protein expression and gene amplification (P 〈 0.01 ). HER -2 protein expression and igene amplification were closely related with breast lesion size and histological grade ( P 〈 0.05 ), while being no significant correlation with patient' s age, tumor, axillary lymph node metastasis and clinical stage of ER and PR positive mark state (P 〉 0.05). Conclusion HER - 2 protein expression and gene amplification has close consistency in breast cancer tissue, and blot of them were closely related to tumor differentiation degree.
