PETIA法测定血清胱抑素C方法学的建立

摘要 建立了一种纳米胶乳微球增强透射免疫比浊法(PETIA)测定胱抑素C(Cys C)的新方法。方法为在p H值=8.0的磷酸盐缓冲液中,血清中的胱抑素C与测定试剂中经纳米胶乳微球修饰后的胱抑素C抗体反应后形成抗原-抗体复合物,并形成一定浊度,该浊度与样品中胱抑素C浓度成正比。该法线性范围0-8.00mg/L,批内变异系数(CV)和批间变异系数分别为0.88%-2.44%和1.12%-3.12%,回收率为99.0%-103.6%,与参考方法较具有良好的相关性,线性回归方程和相关系数分别为Y=0.98X+0.13,r=0.9833。用PETIA法测定血清胱抑素C方法简便、灵敏可靠,适合推广应用。 Established a new PETIA method to determine CystatinC in serum. The analysis was based on the fact that the Cystatin C react to the Cystatin C antibody and then antigen -- antibody Complex was produced, the turbidity of antigen - antibody complex was direct ratio to the concentration of serum Cystatin C. The method was liner in the range of 0 - 8. 00mg/L, the within run and between run CV was 0.88% - 2.44% and 1.12% - 3.12% , the recovery was 99.0% - 103.6%. Comparing this methods (Y) with refence method (X), we obtained the regression equations: Y = 0.98X 4-0. 13 ,r =0. 9833. This method was simple, sensitive and suitable for clinical application.
