摘要
[目的]研究测定灰枣c AMP含量的新方法及灰枣采收期对其c AMP含量的影响。[方法]以阿克苏地区灰枣为研究对象,建立了HPLC法测定灰枣中c AMP含量的定量测定方法,并对不同采收时期的灰枣及其制干产品的c AMP含量进行了测定。[结果]试验表明,HPLC测定c AMP含量的色谱条件为:选用Inertsil ODS-3色谱柱(4.6×250×5μm)、以10%甲醇-90%的浓度为0.167 8%甲酸溶液为流动相、柱温30℃、流速0.8 ml/min、检测波长254 nm测定灰枣中的c AMP含量,c AMP浓度在1.0-100.0μg/ml范围内与峰面积具有良好的线性关系,R2=0.999 0,且其精密性高、重现性好、加标回收率高(〉98%,RSD值为1.38%)。灰枣采收时期对鲜枣及其制干产品的c AMP含量有显著影响(达到0.01显著水平)。[结论]阿克苏地区灰枣在10月24日至10月31日期间采收,有利于保持较高的c AMP含量,此期采收的鲜灰枣及其干枣的c AMP含量分别为75.237和294.097μg/g左右。研究可为新疆阿克苏地区红枣产业化发展提供科学参考。
[Objective] To develop new method for determination of cAMP content in Huizao jujube, and study effects of harvest time on cAMP content. [ Method] With Huizao jujube in Aksu area as research object, the HPLC method was established for determination of cAMP content, cAMP content in Huizao jujube and its dried products in different harvest time was determined. [ Result] The results as follows: the appropriate method with HPLC, were using Inertsil ODS-3 column (4.6 × 250 ×5 μm) at 30 ℃ as stationary phase, using solution composed of methanol-0. 167 8% formic acid ( 10: 90)as mobile phase, detection wavelength at 254 nm, and flow rate with 0.8 ml/min. There was a good linear relationship between the concentration of cAMP and area integral value in the range of 1.0 - 100.0 μg/m, R2 =0. 999 0. The average recovery was over 98.0% with RSD = 1.38. There was significant impact at 0.01 significant level between harvest time of Huizao jujube and their content of cAMP in fresh and dry fruits in Aksu Area. [ Conclusion] The appropriate harvest time was on between October 24 and 31 to obtain higher content of cAMP in Aksu Area, which was 75. 237,294. 097 μg/g or so respectively. The study can provide scientific reference for Huizao jujube industrial development in Aksu Area.
