摘要
以普通玉米淀粉为原料,以抗性淀粉制备产率为考察指标,采用复合酶法制备普通玉米抗性淀粉。利用单因素实验确定最适淀粉乳浓度为25%;利用响应面法研究纤维素酶单独作用,利用正交试验研究纤维素酶和普鲁兰酶复合处理对抗性淀粉制备产率的影响,确定最佳酶解工艺:先加入普鲁兰酶20 U/m L,酶解温度45℃、酶解时间26 h、酶解p H值4.8;再加入纤维素酶40 U/g,酶解时间50 min、酶解温度30℃、酶解p H值5.0。经反复验证,复合酶法制备普通玉米抗性淀粉得率为28.1%。
In this study, in order to determine the optimal conditions of resistant starch from maize by complex of different enzymatic methods, based on one-factor-at-a-time experiments, the main parameters were investigated by response surface methodology(RSM). The results by orthogonal test showed that the optimal conditions were as follows: 25% of the starch concentration, pullulanase amount of 20 U/m L starch, pullulanase reaction time 26 h, temperature 45 ℃, p H 4.8; cellulase amount of 40 U/g starch, cellulase reaction time 50 min, temperature 30 ℃ and p H 5.0, the yield of RS from maize reached to 28.1% under the optimum conditions.
