摘要
目的优化白首乌中C21甾体苷类成分的提取及纯化工艺。方法以白首乌C21甾体苷为考察对象,单因素、正交实验优化提取工艺,考察纯化C21甾体苷的大孔吸附树脂的型号、除杂溶剂、洗脱溶剂及洗脱参数。结果确定8倍量的95%乙醇回流提取2次,每次1.5h,选用HPD-100大孔吸附树脂,分别用2BV的水、1BV的20%的乙醇作为除杂溶剂、3BV 70%乙醇富集洗脱,所得固体中C21甾苷的含量达49%。结论此法提取纯化白首乌中C21甾体苷工艺简单、产物纯度高,可为C21甾体苷的进一步研究提供依据。
Objective To optimize the extraction and purification process of C21 steroidal glycosides from Cynanchum auriculatum. Methods The best extraction technology of C21 steroidal glucosides is decided by single factor and orthogonal experiment. The purification technology is studied by macroporous adsorption resin technology from resin type selection,removing wastes solvent,elution solvent and elution parameters. Results The content of C21 steroidal glycoside is higher when Baishouwu is extracted in 95%alcohol and refluxing and extracting for 2 times,each time 1. 5h. HPD- 100 macroporous adsorption resin is used to purify C21 steroidal glycosides. When the sample is added to the top of column,then eluting by 2BV water,1BV 20% alcohol and 3BV 70%alcohol,70% alcohol fraction is gathered and concentrated with 49% content of C21 steroidal glycoside. Conclusion The extraction and purification technology decided by this study is simple,with a higher content of C21 steroidal glycosides. This study can provide theory evidence for further study of C21 steroidal glucosides.
