摘要
目的:探讨不同糖皮质激素受体(GR)亚型α、β的蛋白及mRNA在血管瘤患儿瘤组织中水平与糖皮质激素治疗血管瘤疗效的相关性。方法:选取2013年2月~2015年1月来医院就诊并同意接受糖皮质激素治疗的血管瘤患儿56例作为研究对象,治疗前、后均细针穿刺取血管瘤组织,根据治疗效果分为激素治疗有效组和激素治疗无效组。采用酶联免疫法和实时荧光定量法检测两组患儿瘤组织中GRα、GRβ蛋白及mRNA水平。结果:激素治疗有效组患儿瘤组织中GRα蛋白及mRNA水平在治疗前明显高于激素治疗无效组,两组比较差异有统计学意义(P〈0.01);GRβ蛋白及mRNA水平在治疗前与激素治疗无效组比较差异无统计学意义(P〉0.05)。结论:糖皮质激素治疗血管瘤患儿的疗效与GRα蛋白及mRNA表达水平呈正相关性,与GRβ蛋白及mRNA表达水平无明显相关性,其治疗机制有待于进一步探讨。
Objective: To explore the correlation between the levels of glucocorticoid receptors (GRs) subtypes α, β proteins and mRNA in tumor tissue of children with hemangioma and curative effect of glucocorticoid. Methods: Fifty-six children with hemangioma and treated with glucocorticoid in the hospital from February 2013 to January 2015 were selected as study objects, fine needle aspiration of he- mangioma was conducted before and after treatment, then the children were divided into effective hormone therapy group and ineffective hor- mone therapy group according to the curative effect. ELISA and real-time fluorescence quantitative method were used to detect the levels of GRoα, GRβ proteins and mRNA. Results: Before treatment, GRα protein and mRNA levels in effective hormone therapy group were statisti- cally significantly higher than those in ineffective hormone therapy group ( P〈0. 01 ) ; there was no statistically significant difference in GRβ protein and mRNA levels between effective hormone therapy group and ineffective hormone therapy group (P〉0. 05 ) . Conclusion: The curative effect of glucocorticoid in treatment of hemangioma is positively correlated with GR α protein and mRNA levels, but the curative effect of glucocorticoid in treatment of hemangioma is not correlated with GR β protein and mRNA, the therapeutic mechanism need to be fur- ther discussed.
