HPLC测定复方苦参止痒软膏中苦参碱的含量

Determination of Matrine in Compound Kushen Antipruritic Ointment by HPLC

目的：建立复方苦参止痒软膏中苦参碱的HPLC含量测定方法。方法：ZORBAX NH2色谱柱（250 mm×4.6 mm,5μm）;流动相为乙腈-无水乙醇-3%磷酸水溶液（80∶10∶10）;流速为0.8 m L·min^-1;检测波长为220 nm;柱温为35℃。结果：苦参碱质量浓度在0.012 5-0.600 mg·mL^-1与峰面积呈良好的线性关系,r=0.999 7,平均回收率为99.8%（n=7）,RSD=2.10%。结论：该方法准确、快速、灵敏度高,可用于复方苦参止痒软膏的质量控制。

Objective：To establish an HPLC method for determination of matrine in Compound Kushen antipruritic ointment.Methods：ZORBAX NH2column（ 250 mm × 4.6 mm,5μm） was used with acetonitrile-ethyl alcohol-3% phosphoric acid（ 80∶10∶10） as the mobile phase,the flow rate was 0.8 m L·min^-1,and the detection wavelength was set at 220 nm with temperature at 35℃.Results：Good linearity was shown between the concentration and peak area in the concentration range of0.125-0.600 mg·m L^-1 for the matrine,The average recovery rate was 99.80% and RSD was 2.10%.Conclusion：The method is accurate,rapid,highly sensitive,and can be used for the quality control of Compound Kusheng antipruritic ointment.
