质粒PLXSN-Bcl-2基因重组体对缺血性脑梗死大鼠血管内皮生长因子的影响 被引量:1

摘要 目的探讨质粒PLXSN-Bcl-2基因重组体对缺血性脑梗死大鼠血管内皮生长因子(VEGF)的影响。方法根据改良线栓法将225只雄鼠制成大脑中动脉阻塞(MCAO)2小时模型,随机分为Bcl-2组(注入质粒PLXSN-Bcl-2)、空质粒组(注入质粒PLXSN)和生理盐水组(注入生理盐水),每组各75只,在缺血再灌注不同时间点检测各组雄鼠的VEGF蛋白及Bcl-2蛋白表达水平和脑梗死体积。结果 Bcl-2组大鼠不同时间点脑梗死体积均低于空质粒组和生理盐水组,差异具有显著性(P<0.05);Bcl-2组大鼠不同时间点VEGF蛋白和Bcl-2蛋白阳性表达水平均显著高于空质粒组和生理盐水组,组间比较差异具有显著性(P<0.05)。结论将质粒作为载体,经大鼠侧脑室导入Bcl-2基因,能够显著减少脑梗死体积,促进VEGF蛋白阳性表达,为缺血性脑梗死治疗提供参考依据。 Objective To investigate the effect on the rats with the influence of VEGF by injecting Bcl-2 into their lateral ventricles. Method 225 male rats were made into middle cerebral artery occlusion (MCAO) model of 2 hours, were randomly divided into Bcl-2 group (injected plasmid PLXSN-Bcl-2), empty plasmid group (injected plasmid PLXSN) and saline group (injected saline), each group had 75 rats, in the ischemia reperfusion in different time were detected in male rats with VEGF, Bcl-2 and the volume of cerebral infarction. Result The volume of cerebral infarction in different time points in rats of Bcl-2 group were lower than the blank plasmid group and saline group, there were statistically signiifcant difference between the three groups (P〈0.05);the positive expression of VEGF protein and Bcl-2 protein in Bcl-2 group rats at different time points were signiifcantly higher than that in empty plasmid group and saline group, there were statistically signiifcant difference between the three groups (P〈0.05). Conclusion The plasmid as a vector, the lateral ventricle of the rat Bcl-2 gene expression, can signiifcantly reduce the volume of cerebral infarction, and promote the positive expression of VEGF, provides the reference for the treatment of ischemic cerebral infarction.
