摘要
目的探讨甲钴胺对坐骨神经损伤的修复作用。方法建立SD大鼠坐骨神经钳夹挤压损伤模型,60只大鼠分为实验组(腹腔注射甲钴胺)、对照组和正常组。每周测量计算坐骨神经功能指数(SFI),术后第4、8周对大鼠进行坐骨神经传导速度检查,取标本测量腓肠肌恢复率,取坐骨神经干进行苏木精-伊红染色,显微镜及电镜观察组织形态学改变。结果实验组SFI较对照组恢复快(P〈0.05)。术后第4、8周,神经电生理与肌肉湿重检测均显示正常组最好(P〈0.05),实验组次之,对照组最差(P〈0.05)。组织学观察显示,第4周时实验组、对照组均出现典型的神经损伤后修复改变;第8周时实验组恢复更接近正常组,其效果明显优于对照组。结论甲钴胺可加快坐骨神经损伤的修复。
Objective To investigate the effects of mecobalamin on sciatic nerve injury. Methods In this experiment, a model of sciatic nerve injury by compression was established in rats. The rats in the treatment group were intraperitoneally injected with mecobalamin. Meanwhile, a normal group and a control group were set. Then, the sciatic functional index (SFI) of each group was measured weekly. The conduction velocities of sciatic nerve in rats were examined on Weeks 4 and 8. Meanwhile, the recovery rates of gastrocnemius muscle were measured, and morphological changes in the tissues were observed under an electron microscope by hematoxylin - eosin staining. Results The treatment group showed faster recovery than the control based on the changes in SFI. The best results of neurophysiological and muscle wet weight detections were found in the normal group ( P 〈 0.05 ) , followed by the treatment group and then the control ( P 〈 0.05 ). Histological observation indicated that the recovery within injured nerves in both treatment and control group on Week 4, while the treatment group presented remarkably improved condition than the control on Week 8, which is more similar to the normal. Conclusion Mecobalamin can accelerate the recovery of sciatic nerve after injury.
