摘要
目的:研究茱杞偏瘫颗粒对脑缺血模型大鼠的保护作用。方法:采用线栓法复制大鼠脑缺血模型。60只大鼠随机均分为假手术(等容纯净水)组、模型(等容纯净水)组、尼莫地平(阳性对照,0.009 g/kg)组与茱杞偏瘫颗粒高、中、低剂量(10.8、5.4、2.7g/kg)组,复制模型前7 d ig给药,每天1次,连续10 d。对大鼠行为学进行测定,测定大鼠脑梗死比例,采用显微图像分析系统观察大鼠脑组织病理形态。结果:与假手术组比较,模型组大鼠神经活动评分升高,横木行走实验评分升高,前肢握力减弱,脑梗死比例升高,差异有统计学意义(P<0.01);脑神经细胞大片消失,坏死区边缘可见神经细胞变性,缺血周围区锥体细胞明显减少,胞体变小,尼氏小体消失,核固缩深染。与模型组比较,茱杞偏瘫颗粒高剂量组大鼠6 h神经活动评分降低,茱杞偏瘫颗粒中剂量组大鼠6、24、48 h内神经活动评分降低,茱杞偏瘫颗粒高、中、低剂量组大鼠横木行走实验评分降低、前肢握力增加,茱杞偏瘫颗粒高、中剂量组大鼠脑梗死比例降低,差异均有统计学意义(P<0.01或P<0.05);茱杞偏瘫颗粒高、中、低剂量组大鼠坏死脑神经细胞减少,锥体细胞形态结构趋于正常,水肿较轻,细胞核较规则。结论:茱杞偏瘫颗粒对脑缺血模型大鼠具有一定的保护作用,可改善其神经活动、横木行走及前肢握力,降低脑梗死比例。
OBJECTIVE: To study the protective effects of Zhuqi piantan granules on the model rats with cerebral ischemia. METHODS : The suture-occluded method was adopted to establish the rat models of cerebral ischernia. 60 rats were randomly divid- ed into sham-operation group (isometric purified water), model group (isometric purified water), nimodipine group (positive con- trol, 0.009 g/kg) and Zhuqi piantan granules high, medium and low doses groups (10.8, 5.4 and 2.7 g/kg). The drugs were given to the rats 7 d before the establishment of the models, ig, once a day, for consecutive 10 d. The behavioristics test was measured. The proportion of the rats with cerebral infarction was determined. Microscopic image analysis system was adopted to make patho- morphological observation on the rats' brains. RESULTS: Compared with sham-operation group, the scores of neural activity in model group were increased and the scores of balance test were increased, grip strength was decreased and the proportion of cere- bral infarction was increased, with significant difference (P〈0.01). For the rats therein, a large number of neurons in the brains was disappeared, nerve cell degeneration was noted in the margin of necrotic area, and the pyramidal cells around the ischemic ar- ea was obviously reduced and became smaller, in which the nissl bodies was disappeared, and karyopyknosis and hyperchromatic nuclei were present. Compared with model group, the 6 h behavioral scores in Zhuqi piantan granules high dose group were de- creased and the 6, 24 and 48 h neural activity scores in Zhuqi piantan granules medium dose group were also decreased; the scores of balance test in Zhuqi piantan granules high, medium and low dose groups were decreased and the grip strength was increased; the proportion of cerebral infarction in Zhuqi piantan granules high and medium dose groups were decreased, with significant differ- ence (P〈0.01 or P〈0.05). Necrotic neurons of the brains in Zhuqi piantan granules high, medium and low dose groups were de- creased, pyramidal cells tended to be normal in shape and structure, edema was mild, and nuclei were almost regular. CONCLU- SIONS: Zhuqi piantan granules have certain protective effects on the model rats with cerebral ischemia, which can improve the neu- rological fimctions, the ability of walking on the beam and the grip strength of rats' forelegs, and reduce the proportion of cerebral ischemia.
