20份棉花品种DNA指纹图谱的构建 被引量:4

Construction of DNA Fingerprinting with SSR Markers for 20 Cotton Cultivars

摘要 选取20份棉花品种,利用SSR分子标记法进行DNA指纹图谱的构建和遗传多样性分析。本研究从92对引物中筛选到15对核心引物,共检测到57种基因型,平均每对引物检测到的基因型数为3.8个,变幅为2-12;引物多态信息含量(PIC)值介于0.7308-0.9571,平均值为0.8782。结果表明,所选的15对引物中,TMB1152是10615-3、新陆早52、ND359-1、新陆早35、Y1169、新陆早36和新陆早47的特异性引物;BNL2646、CGR5565、NAU2238分别是ND359-1、ND359-2和08283、ND012以及10615-3、新陆早36的特异性引物;JESPR157、CIR332分别是新陆早48和Y1169的特异性引物,剩余品种利用引物组合法予以区分,成功建立了20份棉花品种的指纹图谱。 Twenty cotton cultivars were used as materials for the construction of DNA fingerprinting and analysis of genetic diversity with simple sequence repeat (SSR) markers. After screen from 92 candidate primer pairs, a total of 15 SSR primer pairs were selected polymorphic and generated 57 polymorphic loci among the 20 eultivars;Each marker detected 2 - 12 polymorphic loci with an average of 3.8;The polymorphism information content (PIC) ranged from 0. 7308 to 0. 9571, with an average of 0. 8782. The result showed that in the 15 core primer pairs, TMB1152 was specific primer of 10615-3, Xinluzao NO. 52, ND359-1, Xinluzao NO. 35, Y1169, Xinluzao NO. 36 and Xinluzao NO. 47 ; BNL2646 was specific primer of ND359-1, ND359-2 and CGR5565 was specific primer of 08283,ND012;NAU2238 was specific primer of 10615-3, Xinluzao NO. 36;JESPR157 and CIR332 was specific primer of Xinluzao NO. 48 and Y1169 ,respectively ;The remaining varieties were distinguished with primer combination method. The DNA fingerprinting of 20 experimental cotton varieties was successfully established.
