黄芪多糖对放射性肺损伤小鼠模型的TGF-β1、SOD的影响 被引量:17

摘要 目的:研究黄芪多糖对辐照引起的放射性肺损伤小鼠TGF-β1、SOD的影响,以了解黄芪对放射性肺损伤抑制作用机制。方法:近交系C57/BL雌性小鼠120只,随机数字表法分为4组:对照组、单纯照射组、照射+黄芪多糖组(100 mg/kg·d),单纯黄芪多糖组(100 mg/kg·d);每组30只小鼠。照射方式:10 MV-X射线,经前胸单次照射小鼠肺脏中平面13Gy,全肺照射。给药方式:灌胃,照射前30 min给药1次,以后每日给药至实验结束,对照组给予相应同体积生理盐水。在照射后7 d、28 d、56 d处死后予以肺泡灌洗液(BALF)提取计数炎性细胞;另取部分肺组织HE染色,部分肺组织行超氧化物歧化酶(SOD)检测,ELISA法测定血清中TGF-β1。结果:未被照射组别肺组织均无炎症反应。受照射组(单纯照射组、照射+黄芪多糖组)在照射后56 d均表现出不同程度的放射性肺炎病理变化,但照射+黄芪多糖组较单纯照射组炎症反应明显较轻,照射+黄芪多糖组与同时段的单纯照射组比较:肺组织中血清中TGF-β1明显降低,SOD水平明显较高,炎性细胞募集较少。两组间不同时段血清中TGF-β1水平、肺组织中SOD水平以及炎性细胞计数均具有明显差异(P<0.05或P<0.01)。结论:黄芪多糖在放射性肺损伤小鼠模型中通过保护SOD,降低TGF-β1,减轻炎症反应,起到减轻小鼠放射性肺损伤的作用。 Objective: To observe the effect of Astraglus polysaccharide on the level of TGF- β1 and SOD in serum and pathological structure of lung tissue in radiation- induced lung injury mice. Methods: 120 mice C57 / BL( female) were divided into 4 groups: control group,simple irradiation group,irradiation + Astraglus polysaccharide group [100 mg /( kg·d) ],simply Astraglus polysaccharide group [100 mg/( kg·d) ]by random digits table method. There were 30 mice in every groups. Irradiation method: 10MV- X ray,via the anterior chest,whole lung irradiation with 13 Gy by single dose. Drug delivering method: given Astraglus polysaccharide by gavage at 30 minutes before irradiation until the end of the experiment every day,once a day. In the same time,the control group was given saline. After irradiation at 7 d,28 d,56 d,10 mice of each groups were killed. Bronchoalveolar lavage fluid( BALF) was obtained and inflammatory cells were counted. A portion of lung tissue were stained with HE,another part of lung tissues were detected their SOD level. The level of TGF- β1 in serum was detected with ELISA. Results: The lung of control group and simply Astraglus polysaccharide group showed no inflammatory reaction. Different levels of inflammation was found in the lung tissues of the mice of irradiation group and irradiation + Astraglus polysaccharide group in 56 days after radiation. But the inflammation in irradiation +Astraglus polysaccharide group was significantly lighter than in the irradiation group. Compared with the same period,the level of SOD in lung tissue of irradiation + Astraglus polysaccharide group was significantly higher than those of irradiation group. Among the different periods( 7 d,28 d,56 d,after radiation),TGF- β 1 levels of serum in different groups had significant difference( P〈0. 01); SOD levels and inflammatory cells of lung tissue already had significant difference( P〈0. 01 or P〈0. 05). Conclusions: Astraglus polysaccharide can inhibit radiation- induced lung injury in mice by protecting SOD and reducing TGF- β 1 and inflammation.
