粗品肝素钠制备工艺中酶解条件的研究

Study on the enzymatic hydrolysis conditions of the preparation for crude heparin sodium

为了研究胰蛋白酶在粗品肝素钠制备中的酶解条件,通过效价测定,在料液比、酶解时间、酶解温度、酶添加量、盐浓度、酶解液pH 6个单因素实验的基础上,采用正交试验设计来优化工艺参数.实验结果表明,酶解的最佳条件为料液比1∶8,酶解时间3.5h,酶解温度60℃,酶添加量0.1%,盐浓度0.5mol/L,酶解液pH值为8.8.

To explore the best enzymatic hydrolysis conditions of heparin sodium,on the base of liquid ratio, hydrolysis time, hydrolysis temperature, enzyme dosage, salt concentration and enzymatic value of solution by potency determination and orthogonal test design to optimize the process parameters. The results indicate that liquid ratio of 1 ： 8,hydrolysis time 3.5 h, hydrolysis temperature 60 ℃, enzyme dosage of 0.1%, salt concentration 0.5 tool/L, enzy- matic value of solution pHS. 8 are the best conditions of the experiment on hydrolyzing the heparin sodium with trypsin.
