金天格胶囊对大鼠长骨干骨折愈合影响的实验研究 被引量:4

摘要 [目的]研究金天格胶囊对大鼠股骨干骨折愈合的影响,以及应用Micro-CT技术对骨折愈合过程中骨内微结构的观察。[方法]将40只8周龄SD雄性大鼠通过随机数字表法分为两组,分别为实验组和对照组,每组20只。分别建立股骨干闭合骨折髓内钉固定模型,术后第3天始给予相应的干预措施。每组于造模后2周及8周两个时间点各处死实验动物10只。对患肢股骨进行取材后,大体观察骨折愈合情况,通过X线对骨折愈合情况进行评分,利用生物力学实验机测量骨痂的最大载荷、最大载荷恢复率,利用Micro-CT技术计算骨痂的总体积(TV)、骨体积(BV)及骨体积分数(BV/TV%)并对骨痂进行组织形态学观察。[结果]术后2周,实验组和对照组骨折愈合X线评分没有统计学差异。Micro-CT检测结果提示,实验组的TV值高于对照组,但两者的BV及BV/TV%值没有统计学差异。两组骨痂的最大载荷和最大载荷恢复率差异无统计学意义。术后8周,两组的X线评分、TV值没有统计学差异。但实验组的最大载荷、最大载荷恢复率、BV值及BV/TV%值高于对照组,差异有统计学意义。[结论]金天格胶囊可以促进骨折愈合。主要表现在骨折早期它会促进软骨痂的形成,骨折后期加速骨痂的矿化,提高骨折愈合的力学性能。 [Objective] To evaluate the effects of Jintiange capsule in treating fracture healing in rats. [Methods] The 40 Spragu-Dawley rats were divided into two groups randomly. Left femoral shaft fractures were created in all rats closely, and treated with intramedullary nail. The rats of experimental group were administrated with Jintiange capsule. The rats of control group were given water. The rats were sacrificed at 2 and 8 weeks. Parameters of callus in two groups were evaluated by Micro-CT. Endura TEC ELF 3200 mechanical testing machine was used to evaluate the biomechanical effect of fractured femoral shaft. Moreover, the fracture healing was also evaluated by Xray. [Results] At 2 and 8 weeks, there was no significant difference in fracture healing evaluated by X-ray between two groups. The total volume(TV) in callus in experimental group was higher than control group in 2 weeks, but no difference in bone volume(BV), BV/TV and biomechanical property. At 8 weeks, the experimental group was significant in BV, BV/TV and biomechanical property compared with control group, but not in TV. [Conclusion] Jintiange capsule can enhance the fracture healing in rats by increasing callus formation in early phase and improving mineralization in later phase.
