摘要
目的:探讨骨桥蛋白(osteopontin,OPN)与骨涎蛋白(bonesialoprotein,BSP)在非负荷期非埋植型种植体周围骨界面改建中的作用。方法:选用健康雄性杂种犬8只,在其下颌植入非埋植型种植体,分不同时期处死动物。采用免疫组化方法,对不同时期非埋植型种植体-骨界面OPN与BSP进行动态观察,研究种植体-骨界面中OPN与BSP在界面改建过程中的表达变化。结果:种植义齿植入后在非负荷期,随着种植体周围骨组织发生改建,种植体-骨界面表达OPN和BSP有明显规律性,代表其染色强度的灰度值OPN从79.53±2.33增加到122.17±0.65;BSP从79.71±0.55增加到122.82±0.85。1周时界面OPN与BSP都有表达,2周时表达达到高峰,1月时逐渐下降,3月时趋于正常(均P<0.05)。结论:非埋植型种植体在非负荷期,种植体-骨界面OPN与BSP表达的变化有明显的规律性,且OPN与BSP能促进其界面的改建。在维持种植体与骨界面之间的骨性结合形成中起重要作用。
Objective:To investigate the role of osteopontin (OPN) and bonesialoprotein (BSP) in the remodeling bone interface around non-submerged implant during unloaded period. Methods: Eight healthy male mongrel dogs were used to build the animal model. Non-submerged implants were implanted into the bilateral mandibles of the dogs.The dogs were sacrificed in different stages after the procedure.Immunohistochemistry was used to observe the dynamic expression of OPN and BSP of non-submerged implant-bone interface in different stages,so as to study the change in expression of OPN and BSP in remodeling implant-bone interface. Results:According to immunohistochemical study,implant denture remodeled along with bone tissue around the implant after being implanted during unloaded period. The expression of OPN and BSP in the implant-bone interface showed significant regularity.The gray level (representing staining intensity) of OPN was enhanced from 79.53± 2.33 to 122.17±0.65;and that of BSP,from 79.71±0.55 to 122.82±0.85(P〈0.05).The expressions of OPN and BSP were observed after 1 week, peaked after 2 weeks, decreased gradually after 1 month, and dropped to the normal level after 3 months. Conclusion:The expression changes of OPN and BSP in non-submerged implant&amp;nbsp;interface have significant regularity during unloaded period.Besides,OPN and BSP can stimulate the remodeling of the interface,and have an important role in maintaining the formation of bony union between implant and bone interface.
