摘要
[目的]利用分子标记SRAP技术,构建22个甘薯品种的DNA指纹图谱并进行遗传多样性分析。[方法]从49对SRAP引物中筛选了11对引物进行PCR扩增。[结果]共扩增出98条带,多态性条带74条,多态性比率为75.5%,平均每对引物扩增8.91条带。利用引物Me2-em3和Me3-em7,构建了22个甘薯品种的指纹图谱。聚类结果显示,在阈值为15.5时可将22个甘薯品种被分为7类。[结论]地理来源相同的甘薯品种和具有同一亲本的甘薯品种聚在了一起。
[ Objective] By using SRAP maker,22 DNA fingerprints of sweet potato were constructed, also genetic diversity was analyzed. [ Methods] From 49 pairs of SRAP primers, 11 pairs of primers were selected, also they were PCR amplified. [ Results] Total 98 bands were produced,including 74 polymorphic bands, and 75.5% of polymorphic percentage of bands. The average numbers of polymorphic DNA amplified by each primer were 8.91.22 sweet potato species fingerprints were constructed with primer Me2 - em3 and Me3 - em7. The cluster analysis indicated that when threshold is 15.5,22 sweet potato species could be classified into 7 kinds. [ Conclusion] The dendrogram indicated that sweetpotato varieties coming from the same geography places or having same parents were clustered in the same groups.
