摘要
[目的]研究氮源、氮源和矿质元素对菌株MY07产吲哚乙酸量的影响。[方法]在单因素筛选和初始浓度确定试验的基础上,利用响应面分析法对细菌MY07产吲哚乙酸的液体发酵培养基进行优化,采用Box-Benhnken设计和响应面法对碳源、氮源和矿质元素水平进行分析。[结果]结果表明,菌株MY07产IAA的液体发酵培养基最佳组合为:0.94%甘露醇,2.35%KNO3,0.12‰Mg SO4·7H2O,0.13‰Ca Cl2·2H2O,在此条件下的验证试验表明,IAA产量可达到28.16μg/ml。[结论]通过响应面法优化菌株培养基后,有利于提高该菌产生IAA的能力。
[ Objective ] The optimization of liquid fermentation medium for producing IAA by MY07 by response surface methodology. [ Methods] The effects of carbon source, nitrogen source and mineral element on IAA secretion were explored in single factor experi- ments. Based on this, the optimum concetrations of them were determined by Box - Benhnken design combined with response surface analy- sis. [ Results] Mannitol and KNO3 were found to be the best carbon source and nitrogen source, respectively. The optimum concetrations (W/V) of Mannitol,KNO3 ,MgSO4 · 7H20 and CaC12 · 2H20 were to be 0. 94% ,2. 35% ,0. 12%o and 0. 13%o,respectively. The IAA se- cretion in optimized medium reached 28. 16 p,g/ml. [ Conclusion ] The component content of culture medium are optimized by the response surface method, which is beneficial to improve the ability of the MY07 to produce IAA.
