Nesfatin-1对内脏高敏感性大鼠内脏敏感性和结肠动力的影响 被引量:4

The effect of nesfatin-1 on visceral sensitivity and colonic motility of visceral sensitivity rats

摘要 目的探讨nesfatin-1对内脏高敏感大鼠内脏敏感性和结肠动力的影响。方法将新生2 d龄雄性Spragne-Dawley大鼠随机分为空白对照组和模型组,制备内脏高敏感性大鼠模型。模型组分为干预对照组、低浓度干预组、中浓度干预组和高浓度干预组,分别给予侧脑室注射0.5μl 0.9%氯化钠、0.5μmol/L nesfatin-1、5μmol/L nesfatin-1、50μmol/L nesfatin-1,评定各组大鼠内脏敏感性,同时记录各组大鼠结肠电生理指标,对实验结果进行差异性统计分析。结果空白对照组和模型组大鼠体重差异无统计学意义(P>0.05),模型组大鼠较空白对照组易激惹。模型组大鼠内脏敏感性较空白对照组高(P<0.05),同时组织学排除器质性病理改变的存在,提示造模成功。三种不同浓度nesfatin-1干预组大鼠内脏敏感性明显高于干预对照组(P<0.05),且低、中和高浓度干预组之间大鼠内脏敏感性差异亦有统计学意义(P<0.05)。干预对照组大鼠快慢波频率、快慢波最大振幅、收缩波数和收缩波振幅指数均低于低、中和高浓度干预组;低、中浓度干预组快慢波频率和快慢波振幅与高浓度干预组比较,差异有统计学意义,但低浓度干预组与中浓度干预组间收缩波数和收缩波振幅指数差异无统计学意义。结论中枢途径应用nesfatin-1可以影响内脏高敏感性大鼠内脏敏感性和结肠动力。 Objective To explore the effect of nesfatin-1 on visceral sensitivity and colon motility of visceral hypersensitivity rats.Methods Sixty-two days old male Spragne-Dawley rats were divided randomly into control group and model group.The model group was induced to visceral hypersensitive by maternal separation method and acetic acid enema method.The model rats were verified and divided randomly and equally into intervention control group,low concentration intervention group,medium concentration intervention group and high concentration intervention group.Then the visceral sensitivity and colonic electricity activities of different groups,were recorded.Results There was no significant difference between the control group and the model group in weight;the model group rats more irritable than the control group.The model group rats showed higher visceral sensitivity than that control group.In the meantime,the model group rats were excluded organic diseases in histology.The two results suggested model rats were successfully set up.The intervention group rats showed higher visceral sensitivity than that intervention control group;the difference of visceral sensitivity was statistically significant among low concentration intervention group,medium concentration intervention group and high concentration intervention group rats.The intervention control group showed lower volatility,maximum amplitude of fast wave and slow wave,contraction wave and index of contraction than those in low concentration intervention group,medium concentration intervention group and high concentration intervention group.The difference of volatility,maximum amplitude of fast wave and slow wave were statistically significant different between low concentration intervention group or medium concentration intervention group and the high concentration intervention group.The difference of volatility,maximum amplitude of fast wave and slow wave were not significantly different between low concentration intervention group and medium concentration intervention group.Conclusion Nesfatin-1 can affect the visceral sensitivity and colon motility in visceral hypersensitivity rats by central pathway.
