NOD1及COX-2在脂肪组织中表达与妊娠期糖尿病相关性研究 被引量:1

Correlation study of NOD1 and COX-2 expression in adipose tissues with gestational diabetes mellitus

摘要 目的检测核苷酸结合寡聚化结构域1(NOD1)和环氧合酶2(COX-2)在妊娠糖尿病患者脂肪组织中的表达,揭示NOD1和COX-2在妊娠糖尿病发病机制中的意义。方法应用定量逆转录聚合酶链反应(q RT-PCR)和蛋白印迹(Western Blot)技术检测体质指数(BMI)匹配的80例正常葡萄糖耐量孕妇(NGT组)和80例妊娠糖尿病者(GDM组)剖宫产后脂肪组织中NOD1基因m RNA和COX-2蛋白水平。结果 GDM组NOD1 m RNA相对表达量为(3.148±1.037),明显高于NGT组的(0.975±0.296),差异有统计学意义(P<0.01)。GDM组脂肪组织中COX-2蛋白表达水平(1.672±0.452)较NGT组(0.768±0.272)显著升高(P<0.01),NOD1和COX-2表达升高与糖尿病胰岛素抵抗具有密切相关性(P<0.01)。结论脂肪组织中NOD1和COX-2表达的升高可能是GDM患者胰岛素抵抗的重要原因,在GDM发病中起到关键作用。 Objective To observe the m RNA and protein expression levels of nucleotide-binding oligomerization domain(NOD1) and cycloxygenase-2(COX-2) in the adipose tissues for patients with gestational diabetes mellitus and reveal the role of NOD1 and COX-2 in the pathogenesis of gestational diabetes mellitus(GDM).Methods Quantitative RT-PCR(q RT-PCR) and western blot were carried out to detect the expression levels of NOD1 and COX-2 for BMI matched 80 NGT pregnant women and 80 GDM women.Results The relative m RNA expression of NOD1(3.148±1.037) in GDM group were significantly higher than that of NGT group(0.975 ±0.296,P 〈0.01).Protein expression of COX-2 for GDM group(1.672 ±0.452) was remarkably higher than that of NGT group(0.768±0.272,P〈0.01).NOD1 and COX-2 overexpression was closely related with insulin resistance.Conclusion The overexpression of NOD1 and COX-2 in GDM group is closely related with insulin resistance and may play a key role in the pathogenesis of GDM.
