摘要
以雪山鸡为试验材料,通过MALDI-TOF-MS技术检测雪山鸡GARS-GART-AIRS基因候选SNPs位点多态性,分析GARS-GART-AIRS基因的遗传效应,并对其与肌苷酸含量进行关联分析。结果表明:GARS-GART-AIRS基因SNP1、SNP2与胸肌肌苷酸含量呈显著相关,SNP1中CC型个体胸肌肌苷酸含量显著高于AC型和AA型个体,AC型与AA型个体差异不显著;SNP2中TT型个体胸肌肌苷酸含量极显著高于CT型和CC型个体,CT型与CC型个体差异不显著。腿肌肌苷酸含量与胸肌肌苷酸含量变化趋势一致,初步认为这些SNPs应是肌肉肌苷酸含量的主效QTL。
The aim of the present study was to detect the polymorphism of candidate SNPs of GARS-GART-AIRS gene and analyze its genetic effects on inosinc acid and inosinemonpho sphate (IMP) content. Two SNPs of GARS-GART-AIRS associated with IMP content in muscle tissues were scanned by MALDI-TOF-MS to verify the presence of reported SNPs and analyze their genetic effects in Xueshan chickens. The result showed that SNP1, SNP2 of GARS-GART-AIRS had significant correlation with the IMP of the chest muscle (P〈0. 05). The chest muscle content of CC genotype birds SNP1 was higher than that of AC and AA (P〈0.05), while there was no significant differences between AC and AA. In same tissue, SNP2, TT was higher than CT and CC (P〈0. 01), while CT and CC had no significant difference. The IMP content of leg musle and chest had the same trend. These SNPs can be considered as major QTL of inosine monophosphate in muscle.
