血必净联合缺血预处理对大鼠肝缺血再灌注损伤的作用 被引量:6

摘要 目的:探讨血必净联合缺血预处理(IPC)对大鼠肝缺血再灌注损伤(HIRI)的作用。方法180只雄性SD大鼠随机分为五组:假手术组( S组)、缺血再灌注组( IR组)、血必净预处理组( XBJ组)、缺血预处理组( IPC组)和血必净联合缺血预处理组( XBJ+IPC组),每组随机分为三个时间点(6、12及24 h)。参照Pringle法建立大鼠肝脏缺血再灌注模型,分别于肝脏缺血30 min及再灌注6、12与24 h后处理动物,检测血清ALT及AST浓度,光镜下观察肝组织病理学改变、细胞凋亡及血红素氧合酶-1( HO-1)的表达。结果 IR、XBJ、IPC及XBJ+IPC组血清ALT、AST浓度均明显高于S组(P<0.01或P<0.05,24 h点XBJ+IPC组血清AST浓度除外), XBJ及IPC组明显低于IR组(P<0.01),XBJ+IPC组明显低于XBJ、IPC组(P<0.01),XBJ组AST浓度明显低于IPC组(P<0.01)。病理学评分IR、XBJ、IPC及XBJ+IPC组明显高于S组(P<0.01,24 h点XBJ+IPC组除外),XBJ及IPC组明显低于IR组(P<0.01),XBJ+IPC组明显低于XBJ、IPC组( P<0.01)。细胞凋亡指数IR、XBJ、IPC及XBJ+IPC组明显高于S组( P<0.01),XBJ及IPC组明显低于IR组(P<0.01),XBJ+IPC组明显低于XBJ、IPC组(P<0.01)。HO-1评分IR组明显高于S组(P<0.01),XBJ、IPC组明显高于IR组(P<0.01),XBJ+IPC组明显高于XBJ、IPC组(P<0.01)。结论血必净及IPC对大鼠HIRI均有保护作用,两者联合应用效果更好。 Objective To investigate the effect of Xuebijing combined with ischemic preconditioning on liver ischemic-reperfusion injury in rats.Methods One hundred and eighty male SD rats were randomly divided into five groups: sham -operation group ( group S ) , ischemia -reperfusion group ( group IR) , Xuebijing preconditioning group ( group XBJ) , ischemic preconditioning group (group IP) and Xuebijing combined with ischemic preconditioning group (group XBJ+IP) and each group was randomly divided into three time points (6, 12, 24 h).Ischemia-reperfusion model were established according to Pringle's method.After 30 min ischemia and 6, 12, 24 h reperfusion of the liver, the concentrations of ALT and AST in serum were measured, the liver histological changes, cell apoptosis and expression of heme oxygenase-1 were observed under optical microscope.Results At the same time point, serum concentrations of ALT and AST in group IR, XBJ, IP and XBJ+IP were higher than those in group S (P〈0.01 or P〈0.05, except the AST of group XBJ+IP at 24 h);group XBJ and IP were significantly lower than those in group IR ( P 〈0.01 ); group XBJ +IP were significantly lower than those in group XBJ and IP ( P〈0.01); the serum concentrations of AST in group XBJ was significantly lower than those in group IP (P〈0.01).Pathological score in IR, XBJ, IP and XBJ+IP group was higher than that in group S ( P〈0.01, except the XBJ+IPC group at 24 h);group XBJ and IP was significantly lower than that in group IR ( P 〈0.01 ); group XBJ +IP was significantly lower than that in group XBJ and IP (P〈0.01).The apoptotic index in group IR, XBJ, IP and XBJ+IP was significantly higher than that in group S;group XBJ and IP was significantly lower than that in group IR (P〈0.01), group XBJ +IP was significantly lower than that in group XBJ and IP (P〈0.01).The HO-1 score in group IR was significantly higher than that in group S (P〈0.01);group XBJ and IP was significantly higher than that in group IR ( P 〈0.01 ); group XBJ +IP was significantly higher than that in group XBJ and IP (P〈0.01).Conclusion Both of Xuebijing and ischemic preconditioning can protect liver from ischemia reperfusion injury in rats to some extent, and the combined application gives better effects.
