摘要
为研究新城疫病毒(Newcastle disease virus,NDV)的遗传变异特性,对NDV HB/1/05/Dk株的血凝素-神经氨酸酶(haemagglutinin-neuraminidase protein,HN)蛋白基因进行了扩增、测序和遗传进化分析。结果表明:该病毒的HN基因长2 002bp,含有1个长为1 716bp的完整开放阅读框架(Open reading frame,ORF),编码571个氨基酸,符合强毒株的特征。同源性分析表明,该毒株的HN基因与35株参考毒株间的核苷酸和氨基酸的同源性分别为71.8%-99.1%和83.4%-98.9%。遗传进化分析表明,该毒株与基因VIId亚型参考毒株关系较近,属同一分支,与疫苗株B1、LaSota、Mukteswar株和I类毒株关系较远。因此,须加强河北省水禽NDV的监测。
In order to study the genetic variation of Newcastle disease virus (NDV) ,the haemagglutininneuraminidase(HN)protein gene of NDV HB/1/05/Dk isolated from Hebei province was amplified ,sequenced and genetic analyzed .The full length of H N gene comprised of 2002 nucleotides ,encoding 571 amino acids ,w hich is a typical sequence belonging to velogenic strain .The homology analysis of the nucleotide sequences and amino acids of H N gene indicated the isolate had homologies of 71 .8% to 99 .1% and 83 .4% to 98 .9% with 35 strains . Phylogenetic analysis based on the nucleotide sequences of H N gene showed that the HB/1/05/Dk strain was grouped into the genotype VIId virus ,and had a distant genetic relationship with the vaccine strain e .g .B1 ,LaSota ,Mukteswar ,and strains of ClassⅠ .It is necessary to strengthen the monitoring of waterfowl NDV in Hebei province .
