家蚕5龄幼虫农药中毒后中肠和丝腺组织中的部分金属元素含量变化

Variation of Some Metallic Elem ent Contents in Midgut and Silk Gland of the Fifth-instar Silkworm Larvae After Pesticide Poisoning

摘要 蚕体内的金属元素是细胞和组织生理活动不可或缺的基本成分,维持其平衡对家蚕的正常生长发育非常重要。为了解家蚕农药中毒后体内金属元素含量的变化规律,分别给5龄幼虫添食不同浓度的辛硫磷、溴虫腈药液后,运用电感耦合等离子体质谱法(ICP-MS)对幼虫中肠、丝腺组织中的金属元素Ni、K、Na、Rb和Fe进行定量分析。用ICP-MS法检测各金属元素含量在一定质量浓度范围内呈良好的线性关系,相关系数在0.998 0~0.999 9之间。研究结果表明,家蚕5龄幼虫添食辛硫磷和溴虫腈药液后,中肠和丝腺组织中Na、K元素平衡发生变化;由辛硫磷导致的中毒会使幼虫2个组织中的Ni元素含量降低,而溴虫腈中毒会使幼虫2个组织中的Rb元素含量降低。家蚕幼虫中肠和丝腺组织中上述4种金属元素含量的变化,对家蚕农药中毒的生理生化机制研究有重要参考价值。 The metallic elements are indispensable constituents of physiological activity of silkworm (Bombyx mori). Maintaining the balance of metallic element contents in silkworm tissues is very important for their normal growth and de- velopment. In order to understand the variation regularity of metallic element contents in silkworm after pesticide poison- ing, the contents of Ni, K, Na, Rb and Fe in the fifth-instar silkworm larvae after feeding on different concentrations of phoxim-and chlorfenapyr-treated mulberry leaves were determined by inductively coupled plasma-mass spectrometry (ICP-MS). ICP-MS result showed that the contents of various metallic elements presented a good linear relation within a certain concentration range. The correlation coefficients were in the range of 0. 998 0 to 0. 999 9. The results demonstra- ted that the sodium-potassium balance was changed after Phoxim poisoning could lead to the decrease of Ni content the silkworm larvae were fed with phoxim and chlorfenapyr. n the two tissues of silkworm larvae, while chlorfenapyr poisoning could lead to the decrease of Rb content. The analysis of content changes of these 4 mentallic elements provides useful information for the study of physiological and bio- chemical mechanism of silkworm pesticide poisoning.
