摘要
目的:探讨僵蚕天南星方对急性缺血性脑水肿大鼠水通道蛋白4(AQP4)表达的影响及其作用机制。方法:将大鼠随机分为3组:假手术组、模型组、僵蚕天南星方组,采用线栓法制备大鼠大脑中动脉缺血再灌注模型,观察缺血2 h再灌注24 h、48 h、72 h、96 h 4个不同时间点,采用Zausinger评分法观察大鼠神经功能改变,干湿重法测定脑组织含水量,HE染色观察脑组织病理学变化,免疫组织化学法观察大鼠大脑皮层AQP4的表达情况。结果:与模型组比较,僵蚕天南星方组神经功能评分较高(P<0.01),脑组织缺血性病理学损伤均明显改善,脑含水量降低(P<0.01),AQP4蛋白表达量减少(P<0.05)。结论:僵蚕天南星方能够改善脑缺血-再灌注损伤大鼠的神经功能缺损,可减轻脑缺血损伤引起的脑水肿,其机制可能与降低AQP4蛋白的表达有关。
Objective: To investigate the effects of Silkworm Arisaema Decoction on the expression of aquaporin 4(AQP4) and its mechanism of action in rats with acute cerebral edema. Method: The SD rats were randomly divided into three groups:the sham operation group, the model group, the Silkworm Arisaema Decoction group. The model of middle cerebral artery occlu- sion was established by thread ligation method. Adult SD rats were observed cerebral ischemia for 2 h, and reperfusion at 24 h, 48 h, 72 h, 96 h respectively. The Zausinger score was used to evaluate the change of neurological deficit. The brain water content was determined by dry wet weight method. Histopathological changes were observed by HE staining. The expression of AQP4 in cerebral cortex of rats was evaluated by immunohistochemistry. Results: Compared with the model group, the score of neurological function of the Silkworm Arisaema Decoction group was higher(P〈0.01), the pathological injury of brain tissue of the Silkworm Arisaema Decoction group improved significantly, and the brain water content decreased(P〈0.01), the expression of AQP4 was significantly lower in the Silkworm Arisaema Decoction group than that in the model group(P〈0.05).Conclusion: Silk- worm Arisaema Decoction can improve the damage of brain tissue and reduce brain edema for rats with cerebral ischemia-reper- fusion injury which mechanism might be associated with reducing the expression of AQP4.
