摘要
肝纤维化是各种致病因子致肝细胞损伤后的修复过度。肝星状细胞(hepatic stellate cell,HSC)的激活被认为是肝纤维化的核心环节,实验证实多条信号传导通路参与HSC的激活。RNA干扰(RNA interference,RNAi)能有效沉默目的基因,以HSC活化信号通路为切入点,设计小干扰RNA(small interfering RNA,si RNA)抑制相关信号通路的活化,从而达到抑制HSC,激活是目前研究肝纤维化机制的主要手段。该研究综述了RNAi技术在研究HSC活化信号途径中的作用,以求为治疗肝纤维化提供一种新思路。
Hepatic fibrosis is a over-healing response to multiple injuries. Activating hepatic steUate cells is considered to be the central event in liver fibrosis. Several signaling pathways involve HSCs activation. RNA interference technology is a efficient tool being applied in silencing targeted gene expression. Based on the molecular targets of the signaling pathways involving HSCs activation, individual siRNAs are be chosen to block the pathway. We review the effect of RNAi on the signaling pathways involving HSCs activation to come up with an idea treating hepatic fibrosis
